Evaluation of maximal dosing volume for intravitreal injections in cynomolgus monkeys

Intravitreal (IVT) injections are the current method for retinal drug delivery and have been widely used in humans, although it is well known that IVT often cause an increase intraocular pressure (IOP) closely relates to injection volume. Because of anatomical physiological similarities monkey human eye, cynomolgus monkeys preclinical studies on new treatments using large molecules or gene therapy requiring application. However, there only limited information maximal dosing volume monkeys. To determine appropriate injections, comprehensive ocular examinations were conducted IVT-injected eyes Up a 150 µL/eye by injection, no injection-related findings pupillary light reflex gross observations, slit lamp biomicroscopic indirect ophthalmoscopic fundus autofluorescent examinations, pressure, optical coherence tomography anterior posterior segments axial length measurement, electroretinogram waveforms, histopathological treated eyes. leakage solution from eye was observed at µL/eye, indicating technical limitation applicable In addition, recovery IOP elevation immediately after slower than 50 100 µL/eye.
