Enzyme‐based Synthetic Protein Nanoparticles as Colloidal Antioxidants

Protein-based drug delivery systems have gained popularity due to their intrinsic biocompatibility, straightforward surface modification, and potential for therapeutic activity. Among proteins, enzymes are particularly attractive because of specificity, efficient reaction rates, regeneration after substrate turnover, proven track record in the treatment diseases ranging from cancer inherited metabolic lysosomal storage disorders. Herein, we expand upon previous work on electrohydrodynamic jetting by developing a novel class protein nanoparticles that feature enzymes. In particular, report incorporating antioxidant enzyme, catalase, at weight fractions as high 50%. Catalase-based synthetic (CAT-SPNPs) demonstrated sustained antioxidative activity, retained significantly enhanced enzymatic activity compared its solute form, overall good structural stability. Moreover, functionalization CAT-SPNPs with targeting antibodies resulted ∼12.5-fold increase uptake over unmodified control particles. Importantly, exerted protection oxidative stress, indicated significant viability reduction LDH release equivalent amounts free catalase.
