Environmentally Relevant Mixture of Pesticides Affect Mobility and DNA Integrity of Early Life Stages of Rainbow Trout (Oncorhynchus mykiss)

چکیده
The aim of this study was to analyze the impact three concentrations a pesticide mixture on first development stages rainbow trout (Oncorhynchus mykiss). made up commonly used pesticides in viticulture: glyphosate (GLY), chlorpyrifos (CPF) and copper sulfate (Cu). Eyed stage embryos were exposed for 3 weeks mixture. Lethal sub-lethal effects assessed through number phenotypic molecular endpoints including survival, hatching delay, success, biometry, swimming activity, DNA damage (Comet assay), lipid peroxidation (TBARS), protein carbonyl content gene expression. Ten target genes involved antioxidant defenses, repair, mitochondrial metabolism apoptosis analyzed using real-time RT-qPCR. No significant increase mortality, half-hatch, growth defects, TBARS contents observed whatever concentration. In contrast, activity significantly more elevated at highest Gene transcription up-regulated detoxification (gst mt1), repair (ogg1), (cox1 12S), cholinergic system (ache). This highlighted induction adaptive behavioral responses larvae when environmentally realistic pesticides.

Rainbow Trout, Oncorhynchus mykiss
Rainbow trout are classified as Oncorhynchus mykiss, and as such belong to the same genus as Pacific salmon, and to the family Salmonidae, which includes Atlantic salmon (Salmo salar), various trout (Salvelinus sp.), Arctic char (Salvelinus alpinus), Arctic grayling (Thymallus arcticus) and whitefish (Coregonus sp.). Rainbow trout are native to areas around the North Pacific Ocean, from souther...
متن کاملToxicity comparison of colloidal silver nanoparticles in various life stages of rainbow trout (Oncorhynchus mykiss)
Recognizing the significance of the life stage of fish for nano-eco-toxicological studies, the acute toxicity of colloidal silver nanoparticles (AgNPs) was tested in three different life stages of rainbow trout. Fishes were exposed to colloidal AgNPs at nominal concentrations of 100, 32, 10, 3.2, 1, 0.32, 0.1, and 0.032 mg/L. The estimated 96 hr LC50 values were 0.25, 0.71, and 2.16 mg/L for th...

متن کاملInduction of ornithine-urea cycle enzymes and nitrogen metabolism and excretion in rainbow trout (Oncorhynchus mykiss) during early life stages
The ornithineurea cycle (OUC) is present in elasmobranch fish and many terrestrial vertebrates. Recently, a functional OUC has been reported in a few teleost species, suggesting that all teleost fish have the genes for the OUC, but expression is relatively rare. We investigated the possibility that the OUC is expressed during early development in trout as a mechanism for detoxifying ammoni...
متن کاملContinuous exposure to infectious pancreatic necrosis virus during early life stages of rainbow trout, Oncorhynchus mykiss (Walbaum).
Rainbow trout, Oncorhynchus mykiss (Walbaum), were exposed continuously to infectious pancreatic necrosis virus (IPNV) at 0, 10(1), 10(3) or 10(5) plaque forming units (pfu) L(-1) of water to estimate the effects of chronic IPNV exposure on early life stages. Fish density averaged 35 fish L(-1) (low density) or 140 fish L(-1) (high density), and the tank flow rate was 250 mL(-1) min. Virus expo...
