Endothelial cell metabolism: A potential target to improve tumor immunity
نویسندگان
چکیده
Failure of current therapies to eliminate tumors is, in part, due inadequate infiltration immune cells. The importance endothelial cells for the induction responses is now being recognized. However, tumor (TECs) are dysfunctional, leading decreased immunity. Because TEC metabolism contributes abnormal vascular phenotype, interest growing vasculature as a target improve responses. Recent insights into relationship between and immunity suggest that normalization through metabolic reprogramming could be an important tool immune-based anti-cancer therapies.

dothelial progenitor cells (EPC) participate in angiogenesis and vascular homeostasis by incorporating into the endothelium of damaged vessels and, perhaps more importantly, exhibiting potent angiogenic paracrine effects (1). EPC are embedded in the microenvironment of bone marrow stromal and endothelial cells and translocate to the circulation upon NO-mediated signaling (2). Additional mobiliz...
متن کاملTargeting Tumor Metabolism: A New Challenge to Improve Immunotherapy
Currently, a marked number of clinical trials on cancer treatment have revealed the success of immunomodulatory therapies based on immune checkpoint inhibitors that activate tumor-specific T cells. However, the therapeutic efficacy of cancer immunotherapies is only restricted to a small fraction of patients. A deeper understanding of key mechanisms generating an immunosuppressive tumor microenv...
متن کاملTumor Microenvironment Metabolism: A New Checkpoint for Anti-Tumor Immunity
When a T cell infiltrates a tumor, it is subjected to a variety of immunosuppressive and regulatory signals in the microenvironment. However, it is becoming increasingly clear that due to the proliferative and energetically-deregulated nature of tumor cells, T cells also operate at a metabolic disadvantage. The nutrient dearth of the tumor microenvironment (TME) creates "metabolic checkpoints" ...
متن کاملCell Cycle Regulating Kinase Cdk4 as a Potential Target for Tumor Cell Treatment and Tumor Imaging
The cyclin-dependent kinase (Cdk)-cyclin D/retinoblastoma (pRb)/E2F cascade, which controls the G1/S transition of cell cycle, has been found to be altered in many neoplasias. Inhibition of this pathway by using, for example, selective Cdk4 inhibitors has been suggested to be a promising approach for cancer therapy. We hypothesized that appropriately radiolabeled Cdk4 inhibitors are suitable pr...
متن کاملTargeting Colorectal Cancer Cell Lines Using Nanobodies; AgSK1as a Potential Target
Background: Colorectal cancer is the third most common type of aggressive cancers. Chemotherapy, surgery, and radiotherapy are the common therapeutic options for treating this cancer. Due to the adverse side-eff ects of these methods, immunotherapy is considered as an appropriate alternative therapeutic option. Treatment through the application of monoclonal antibodies is considered as a n...
