Emerging Insights into Molecular Mechanisms of Inflammation in Myelodysplastic Syndromes

Inflammation impacts human hematopoiesis across physiologic and pathologic conditions, as signals derived from the bone marrow microenvironment, such pro-inflammatory cytokines chemokines, have been shown to alter hematopoietic stem cell (HSCs) homeostasis. Dysregulated inflammation can skew HSC fate-related decisions, leading aberrant potentially contributing pathogenesis of hematological disorders myelodysplastic syndromes (MDS). Recently, emerging studies used single-cell sequencing muti-omic approaches investigate cellular heterogeneity gene expression in normal well myeloid malignancies. This review summarizes recent reports mechanistically dissecting role inflammatory signaling innate immune response activation due MDS progression. Furthermore, we highlight growing importance using multi-omic techniques, profiling deconvolution methods, unravel MDSs’ heterogeneity. These provided valuable insights into patterns clonal evolution that drive progression elucidated impact on composition microenvironment MDS.
