Electrical characteristics of amyloid beta peptides in vertical junctions

چکیده
Abstract Assembled amyloid beta (Aβ) peptides have been considered pathological assemblies involved in human brain diseases, and the electron transfer or transport characteristics of Aβ are important for formation structured assemblies. Here, we report electrical surface-assembled similar to those observed Alzheimer’s patients. These correlate their characteristics. Electrical current–voltage plots vertical junction devices show sequence dependence current densities at both monomers ( mono -Aβs) oligomers oli -Aβs), while is not clearly planar field effect transistors (FETs). In particular, surface oligomerization drastically decreases activity transfer, which presents a change pathway junctions. Electron oli- junctions symmetric (tunneling/tunneling) due weak voltage-independent coupling less redox-reactive -Aβ contacts, that asymmetric (hopping/tunneling) redox levels voltage-dependently coupled with contact electrodes. Consequently, through junctions, sequence- conformation-dependent Aβs can reveal activities.

P135: The Role of Amyloid Beta-Peptides and Tau Protein in Alzheimer\'s Disease
Alzheimer's desease is the most common age-related neurodegenerative disorder, and cognitive problems such as defects in learning and memory are of its symptoms. Among the factors involved in the pathogenesis of the disease are biochemical disorders in protein production, oxidative stress, decreased acetylcholine secretion and inflammation of the brain tissue. Extra-neuronal accumulation ...
متن کاملMembrane disordering effects of beta-amyloid peptides.
The interaction of Abeta with synaptosomal plasma membranes decreases membrane fluidity. Using model membrane/liposome systems the interaction of Abeta with specific lipids (e.g. phospholipids, gangliosides, cholesterol) has been defined. The formation of the beta-sheet structure of Abeta when undergoing peptide aggregation is important for Abeta's membrane perturbing properties. This effect ca...
متن کاملProtection against beta-amyloid-induced apoptosis by peptides interacting with beta-amyloid.
beta-Amyloid peptide produces apoptosis in neurons at micromolar concentrations, but the mechanism by which beta-amyloid exerts its toxic effect is unknown. The normal biological function of beta-amyloid is also unknown. We used phage display, co-precipitation, and mass spectrometry to examine the protein-protein interactions of beta-amyloid in normal rabbit brain in order to identify the bioch...
متن کاملAmyloid Beta Peptides Differentially Affect Hippocampal Theta Rhythms In Vitro
Soluble amyloid beta peptide (A β ) is responsible for the early cognitive dysfunction observed in Alzheimer's disease. Both cholinergically and glutamatergically induced hippocampal theta rhythms are related to learning and memory, spatial navigation, and spatial memory. However, these two types of theta rhythms are not identical; they are associated with different behaviors and can be differe...
متن کاملCatechol derivatives inhibit the fibril formation of amyloid-beta peptides.
The inhibition of fibril formation of amyloid beta proteins (A beta) would be attractive therapeutic targets for the treatment of Alzheimer's disease (AD). Dopamine (DA) and other catechol derivatives were used as inhibitory factors for A beta fibril formation. The fibril formation of A beta was monitored by Thioflavin T fluorescence, a transmission electron microscopy (TEM) and a total interna...
