Effects of LSD1 Inhibition on Macrophage Specialization into a Pro-Inflammatory Phenotype

Under the influence of many factors, such as cytokines or chemokines, macrophages specialize into two subpopulations: pro-inflammatory M1 (classical pathway) anti-inflammatory M2 (alternative pathway). Upon stimulation with bacterial ligand PAM3CSK4 and upon LPS (Lipopolysaccharide), TLR (toll-like receptors) 1/2 receptors TLR4, respectively, activate NFκB pathway, which leads to downregulation catalase expression through activity LSD1 HDAC1 complex. The main factor responsible for CAT repression is recruitment promoter site gene, resulting in pausing RNA polymerase. Inhibition SP2509 a decreased IL1b COX2, well some surface proteins, e.g., TLR2, despite presence LPS. iLSD1 prevents thereby inhibition macrophage polarization classic phenotype. In conclusion, regulation determines direction specialization.
