Effective DNA extraction method for metagenomic analysis of rhizosphere bacteria from mangrove sediments

Mangrove, Rhizophora mucronata, grows in the intertidal area, which contains much organic matter and varying salinity. The content is influenced by bacterial community that inhabits ecosystem, but information regarding community, especially mangrove root system, not widely available. There are several challenges completing this information, one of method used still a conventional form. Developments environmental DNA analysis can support complement information. However, must be optimized because salt variations affect extraction results. Thus, study aimed to determine optimal approach for extracting from sediments. two methodologies: manual techniques based on buffer modification kits. were four different treatments, namely soil isolation plus kit (M1), fecal / microbial quick-DNA miniprep (M2), glass powder with charcoal (M3), skimmed milk (M4). samples obtained each assessed concentration purity using nanodrop.
