چکیده
Successful embryo implantation and appropriate trophoblast invasion is associated with an adequate communication interaction between peripheral blood mononuclear cells (PBMC) the human embryo. Human chorionic gonadotropin (hCG) one of earliest embryonic signals that affect PBMC cytokine secretion. Nowadays, intrauterine administration hCG-activated autologous a widely used treatment for improvement pregnancy outcome. However, effect hCG on secretion scarcely studied. The aim present study was to estimate activation pro- anti-inflammatory cytokines (TNF-a, IL6, IL4, IL10). Blood samples (8 ml) were collected from 36 women during mid-luteal phase cycle. isolated cultured (2x106 cells/well) in 0.5 ml RPMI-1640 medium (R8758, Sigma-Aldrich, Germany) + 1% serum albumin (HSA) without 10IU/ml (Choriomon, IBSA, Switzerland) 24h. Tumor necrosis factor alpha (TNFα), Interleukin-6 (IL-6), Interleukin-4 (IL-4) Interleukin-10 (IL-10) cell culture media determined by sandwich enzyme-linked immunosorbent assay (ELISA) (CSB-E04638h, CSB-E04740h, CSB-E04633h CSB-E04593h, Cusabio Technology, Wuhan, China, respectively). Changes in some pro-and anti-inflammatory cytokines produced by bovine peripheral blood mononuclear cells following foot and mouth disease vaccination
Interleukin (IL)-17 is exclusively produced by CD4 helper T-cells upon activation. It most often acts as a pro-inflammatory cytokine, which stimulates the release of pro-inflammatory cytokines IL-6, IL-8, TNF-α, and granulocyte-macrophage colony-stimulating factor (GM-CSF). In this study, we studied the in-vitro IL-17 response to specific antigens and a variety of mitogens and compared the IL-1...
متن کاملHCG-Activated Human Peripheral Blood Mononuclear Cells (PBMC) Promote Trophoblast Cell Invasion
Successful embryo implantation and placentation depend on appropriate trophoblast invasion into the maternal endometrial stroma. Human chorionic gonadotropin (hCG) is one of the earliest embryo-derived secreted signals in the peripheral blood mononuclear cells (PBMC) that abundantly expresses hCG receptors. The aims of this study were to estimate the effect of human embryo-secreted hCG on PBMC ...

BACKGROUND The aim of this study was to investigate the role of peripheral blood mononuclear cells (PBMC) in embryo invasion at the implantation site and to estimate the effect on PBMC function of human chorionic gonadotrophin (HCG) that is secreted from the human embryo. METHODS AND RESULTS The effect of PBMC on the invasiveness of murine embryos was examined using an invasion assay. PBMC ob...

Cryopreservation is the method of choice for long term storage of human PBMCs. This study was designed to compare the different combinations of variables affecting the cryopreservation of PBMCs samples. The viability of PBMCs separated from 2×5 ml peripheral blood samples obtained from 16 healthy adult volunteers, were measured using trypan blue dye exclusion method just before freezing with ...
متن کاملEvaluation of in vitro Effect of Pyocyanine Pigment on Interleukin-2 Secretion from Peripheral Blood Mononuclear Cells in Cancer Patients
Background and Objective: The pyocyanine pigment in Pseudomonas aeruginosa stimulates blood cells to secrete IL-2. IL-2 cytokine is an activator of cytotoxic T cells and natural killer cells. These cells destroy the target cells of patients with cancer. This study aimed to evaluate the effects of pyocyanine on the IL-2 secretion from peripheral blood mononuclear cells (PBMCs) in patients with b...
