eDNA metabarcoding fish in relation to the Water Framework Directive indices

The Water Framework Directive (WFD) recognizes fish as a biological quality element to determine the ecological health and of waterbodies. Therefore monitoring is carried out using conventional methods like electrofishing seine fishing following standardized protocols. This data then used in metrics that calculate ratios (EQR) for eDNA-metabarcoding could prove an alternative this WFD-monitoring by offering high sensitivity, possibly lower costs less invasive sampling. A large project was set up Netherlands together with STOWA waterboards explore possibilities use way carry fish. In 2015 total 55 WFD transects (250 meter) were sampled parallel WFD-monitoring. On average 60% mores species detected eDNA-metabarcoding. 2016 9 whole waterbodies (small rivers/ lakes / canals) samples parallel. sample strategy mixed water collected from 3 long compared individually. results showed resulted nearly same at far costs.
