Dual Drug Loaded Nanotheranostic Platforms as a Novel Synergistic Approach to Improve Pancreatic Cancer Treatment

This study focuses on the development of theranostic, dual drug-loaded nanocarriers to propose a proof-of-principle therapeutic approach in treatment pancreatic ductal adenocarcinoma (PDAC). The nanoconstructs consist core zinc oxide nanocrystals doped with gadolinium, useful as potential contrast agent magnetic resonance imaging applications. After functionalizing their surface amino-propyl groups, physical adsorption two hydrophobic drugs is performed: Vismodegib and Sorafenib. Their synergistic use might improve PDAC stroma depletion when co-delivered tumor microenvironment for future vivo To enhance nanoconstructs’ biostability, ensemble coated by lipid bilayer targeting peptide incorporated outer shell surface. As first proof concept, resulting are tested against cancer cell lines, showing modest increase efficacy compared free drug counterparts proving spare healthy cells. In second testing set, dual-drug loaded both lines previously sensitized first-line chemotherapeutic drug, Gemcitabine, an improved response. From these preliminary results, nanotheranostic platforms constitute good starting point therapy diagnosis studies.
