Down-regulation of NRIP1 alleviates pyroptosis in human lens epithelial cells exposed to hydrogen peroxide by inhibiting NF-κB activation

Purpose: To investigate the role of nuclear receptor-interacting protein 1 (NRIP1) in oxidative stressinduced apoptosis and pyroptosis cataract disease.Methods: Human lens epithelial cells (HLE-B3 cells) were exposed to hydrogen peroxide (H2O2). NRIP1 expression (H2O2)-treated HLE-B3 was determined by western blotting quantitative reverse transcription polymerase chain reaction (qRT-PCR). CCK8 EdU staining used assess cell viability. Flow cytometry pyroptosis.Results: significantly up-regulated post-H2O2 incubation (p < 0.01). Hydrogen reduced viability proliferation cells, while knockdown enhanced proliferation. silencing attenuated H2O2-induced increase NLRP3, N-terminal domain gasdermin D, caspase-1, interleukin (IL)-1β, IL-18 HLEB3 but suppressed H2O2-treated cells. down-regulated cytoplasmic NF-κB NF-κB, increased decreased interference.Conclusion: down-regulation represses human inhibiting activation, thus, providing a potential strategy treat disease.
