چکیده

Abstract Background Intestinal dysbiosis is a key feature in the pathogenesis of inflammatory bowel diseases (IBD). Bacterial quorum sensing mediated by acyl-homoserine lactones (AHL) might play role dialogue between gut microbiota and host. The main objective our study was to investigate presence expression AHL synthase receptor genes human ecosystem during IBD. Methods To confirm gut, mass spectrometric detection performed on stool samples from IBD patients non-IBD subjects. Then, an silico approach, we exploited open access database: Inflammatory Bowel Disease Multi'omics Database, American cohort with bacterial metagenomes metatranscriptomes data characterise dysbiosis, most discriminating species were identified multivariate analysis allowed us define two groups (dysbiosis/non-dysbiosis). search for (luxI) (luxR) known homolog genes, using Basic Local Alignment Search Tool (BLAST) previously assembled gene files (presence/absence) as well raw sequencing (relative abundance expression). Results Mass spectrometry confirmed higher concentration molecules healthy subjects than relapsing Regarding analysis, 103 selected including 50 Crohn's disease (CD), 27 ulcerative colitis (UC), 26 No luxI-like retrieved BLAST searches. However, several homologs identified: sdiA Escherichia coli (7/103 patients) luxR-like Bacteroides fragilis dorei present all patients. According disease, only one under-expressed (p = 0.02) compared non-IBD, especially CD (Figure 1). In situation, luxR appeared be over-expressed 0.04) non-dysbiotic 2). Conclusion Through this computational AHL-synthesising bacteria have not been found. appears modulated IBD-associated dysbiosis. LuxR receptors, species, should investigated understand its impact 3). Acyl homoserine-lactone quorum-sensing signal generation.

Acyl homoserine lactones (acyl-HSLs) are important intercellular signaling molecules used by many bacteria to monitor their population density in quorum-sensing control of gene expression. These signals are synthesized by members of the LuxI family of proteins. To understand the mechanism of acyl-HSL synthesis we have purified the Pseudomonas aeruginosa RhlI protein and analyzed the kinetics of...

Acyl-Homoserine Lactone Quorum Sensing in the Roseobacter Clade

Members of the Roseobacter clade are ecologically important and numerically abundant in coastal environments and can associate with marine invertebrates and nutrient-rich marine snow or organic particles, on which quorum sensing (QS) may play an important role. In this review, we summarize current research progress on roseobacterial acyl-homoserine lactone-based QS, particularly focusing on thr...

Production of acyl-homoserine lactone quorum-sensing signals by gram-negative plant-associated bacteria.

Many gram-negative bacteria regulate expression of specialized gene sets in response to population density. This regulatory mechanism, called autoinduction or quorum-sensing, is based on the production by the bacteria of a small, diffusible signal molecule called the autoinducer. In the most well-studied systems the autoinducers are N-acylated derivatives of L-homoserine lactone (acyl-HSL). Sig...

Structural basis and specificity of acyl-homoserine lactone signal production in bacterial quorum sensing.

Synthesis and detection of acyl-homoserine lactones (AHLs) enables many gram-negative bacteria to engage in quorum sensing, an intercellular signaling mechanism that activates differentiation to virulent and biofilm lifestyles. The AHL synthases catalyze acylation of S-adenosyl-L-methionine by acyl-acyl carrier protein and lactonization of the methionine moiety to give AHLs. The crystal structu...

INFLAMMATORY BOWEL DISEASE Gut-associated bacterial microbiota in paediatric patients with inflammatory bowel disease

Background: Clinical and experimental observations in animal models indicate that intestinal commensal bacteria are involved in the initiation and amplification of inflammatory bowel disease (IBD). No paediatric reports are available on intestinal endogenous microflora in IBD. Aims: To investigate and characterise the predominant composition of the mucosa-associated intestinal microflora in col...
