چکیده

The clustered regularly interspaced short palindromic repeats (CRISPR)/associated protein (CRISPR/Cas) system has been exploited as an efficient gene editing tool with precise site specificity and high efficiency; the delivery of CRISPR/Cas is critical for efficacy therapeutics remains challenging. DNA nanomaterials are widely utilized promising carriers agents because their excellent molecular recognition capabilities, sequence programmability, biocompatibility. Herein, recent advances in systems summarized. Based on construction strategy, nanomaterial-based categorized branched DNA-based nanostructures or rolling circle amplification (RCA)-based nanostructures. Representative studies design highlighted. current challenges opportunities development also discussed. It envisioned that nanotechnology, will open up new possibilities CRISPR/Cas-based therapeutics.

Function and Regulation of Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) / CRISPR Associated (Cas) Systems

Phages are the most abundant biological entities on earth and pose a constant challenge to their bacterial hosts. Thus, bacteria have evolved numerous 'innate' mechanisms of defense against phage, such as abortive infection or restriction/modification systems. In contrast, the clustered regularly interspaced short palindromic repeats (CRISPR) systems provide acquired, yet heritable, sequence-sp...

Non-classical phase diagram for virus bacterial coevolution mediated by clustered regularly interspaced short palindromic repeats.

CRISPR is a newly discovered prokaryotic immune system. Bacteria and archaea with this system incorporate genetic material from invading viruses into their genomes, providing protection against future infection by similar viruses. The condition for coexistence of prokaryots and viruses is an interesting problem in evolutionary biology. In this work, we show an intriguing phase diagram of the vi...

Experimental definition of a clustered regularly interspaced short palindromic duplicon in Escherichia coli.

We analyzed sequences of newly inserted repeats in an Escherichia coli CRISPR (clustered regularly interspaced short palindromic repeats) array in vivo and showed that a base previously thought to belong to the repeat is actually derived from a protospacer. Based on further experimental results, we propose to use the term "duplicon" for a repeated sequence in a CRISPR array that serves as a tem...

Insight into Microevolution of Yersinia pestis by Clustered Regularly Interspaced Short Palindromic Repeats

BACKGROUND Yersinia pestis, the pathogen of plague, has greatly influenced human history on a global scale. Clustered Regularly Interspaced Short Palindromic Repeat (CRISPR), an element participating in immunity against phages' invasion, is composed of short repeated sequences separated by unique spacers and provides the basis of the spoligotyping technology. In the present research, three CRIS...

Curbing Malaria: A New Hope through Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) Technology

The term genome refers to the total genetic composition of an organism or species. The normal genes transcribe and translate into functional gene products, which in turn bring about a normal phenotype. Genomics, the study of genomes has revealed the significant level of diversity in the human genome attributed not only to single nucleotide polymorphisms (SNPs) but structural variations as well....
