چکیده

Since 2004, unspecific peroxygenases, in short UPOs (EC. 1.11.2.1), have been explored. are closing a gap between P450 monooxygenases and chloroperoxidases. These enzymes highly active biocatalysts for the selective oxyfunctionalisation of C–H, C=C C-C bonds. secreted fungal proteins Komagataella phaffii (Pichia pastoris) is an ideal host high throughput screening approaches UPO production. Heterologous overexpression 26 new by K. was performed deep well plate cultivation shake flask up to 50 mL volume. Enzymes were screened using colorimetric assays with 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), 2,6-dimethoxyphenol (DMP), naphthalene 5-nitro-1,3-benzodioxole (NBD) as reporter substrates. The PaDa-I (AaeUPO mutant) HspUPO used benchmarks find interesting complementary activity profiles good producing strains. Herein we show that six from Psathyrella aberdarensis, Coprinopsis marcescibilis, Aspergillus novoparasiticus, Dendrothele bispora brasiliensis particularly active.

Fungal Unspecific Peroxygenases: a New Generation of Oxygen-transferring Biocatalysts

The oxygenation of organic molecules is a challenging task in synthetic chemistry and therefore biocatalytic approaches using oxygen-transferring enzymes have come into the focus of chemists and biotechnologists. Fungal peroxygenases represent a unique enzyme type that selectively transfers oxygen from peroxides (R-OOH) to numerous substrates such as benzene derivatives, polycyclic aromatic hyd...

Fungal Unspecific Peroxygenases Oxidize the Majority of Organic EPA Priority Pollutants

Unspecific peroxygenases (UPOs) are secreted fungal enzymes with promiscuity for oxygen transfer and oxidation reactions. Functionally, they represent hybrids of P450 monooxygenases and heme peroxidases; phylogenetically they belong to the family of heme-thiolate peroxidases. Two UPOs from the basidiomycetous fungi Agrocybe aegerita (AaeUPO) and Marasmius rotula (MroUPO) converted 35 out of 40 ...

Synthesis of novel lipids in Saccharomyces cerevisiae by heterologous expression of an unspecific bacterial acyltransferase.

The bifunctional wax ester synthase/acyl-coenzyme A:diacylglycerol acyltransferase (WS/DGAT) is the key enzyme in storage lipid accumulation in the gram-negative bacterium Acinetobacter calcoaceticus ADP1, mediating wax ester, and to a lesser extent, triacylglycerol (TAG) biosynthesis. Saccharomyces cerevisiae accumulates TAGs and steryl esters as storage lipids. Four genes encoding a DGAT (Dga...

Heterologous Expression of Bovine Prochymosin in Pichia pastoris GS115

Objectives: In present research we evaluate the expression of this critical enzyme in a eukaryotic system for future use in cheese industry. Materials and Methods: We have cloned bovine prochymosin gene in methylotrophic yeast, P. pastoris, using pPIC9K as an expression vector. The recombinant plasmid was transformed into the host by electroporation, and it was expre...

Heterologous Expression of Potato Virus Y Coat Protein, Isolate Pot187

Background: The advent of recombinant DNA technology has facilitated heterologous expression of proteins from various sources in different host systems including Escherichia coli. If a plant virus coat protein is expressed in the bacterium it can be used as the antigen for antibody preparation. Such a recombinant antigen preparation can be particularly useful where equipment such as ultracentri...
