Background: Fluid cytology plays an important role in delineating benign from malignant effusions, tumor staging, and also diagnosing recurrences. Various methods are used for the preparation of smears. As accurate diagnosis fluids aids clinical decisions, method smears, it is very important. Cytospin smears one which provide higher cellular yield with better preservation morphology less time consuming. On other hand, cell block gives superior architectural details provides options immunocytochemistry. Aims Objectives: The aim this study was to assess diagnostic utility cytospin comparison peritoneal pleural fluid cytology. done determine sensitivity, specificity, predictive values, accuracy preparations considered as gold standard. Materials Methods: This a test evaluation at Department Pathology, Government Medical College, Kottayam. sample size 240 included all received our laboratory during period. prepared were compared tissue sections by method, evaluate cytospin. Tissue blocks effusions standard comparison. Results: A preparations. sensitivity cytopsin 94%. specificity 100%. positive value 100% negative 96.8%. Hence, 97.9%. Conclusion: There only minimal statistical difference between results obtained methods. consuming along advantage yield. incorporation technique beneficial augmenting effusion
