چکیده
The aim of the present study was to investigate mechanism by which dexmedetomidine (DEX) alleviates neuropathic pain in a chronic constriction injury (CCI) model rats. A CCI rat established through sciatic nerve ligation. rats were treated with DEX, nuclear factor erythroid 2?related 2 (Nrf2) inhibitor ML385, NLR family pyrin domain containing 3 (NLRP3) antagonist MCC950 and/or NLRP3 activator nigericin. mechanical withdrawal threshold (MWT) measured assess sensitivity Hematoxylin and eosin staining TUNEL used examine spinal apoptosis, respectively. ELISA quantify levels inflammatory factors. expression Nrf2 also examined. results indicated that decrease MWT increases cord injury, apoptosis factors detected compared control Spinal inflammation abrogated DEX?treated Compared group, an increase decreases MCC950, while opposite effects observed on these indicators DEX + ML385 groups groups, increased, decreased nigericin group group. In summary, reduced suppressing activation.

Curcumin Alleviates Streptozotocin-Induced Hyperalgesia in Diabetic Neuropathic Rat via Attenuation of Lipid Peroxidation and Inflammation
Background and Objective: Hyperalgesia is one of the marked clinical signs of patients with diabetes mellitus that affects their lifestyle. This study was conducted to assess the anti-nociceptive effect of chronic administration of curcumin in streptozotocin (STZ)-diabetic rats using formalin, hot tail immersion, and paw pressure tests. Materials and Methods: This study was conducted on 40 male...

متن کاملSodium butyrate alleviates adipocyte inflammation by inhibiting NLRP3 pathway
Insulin resistance (IR) is a common feature of Type II diabetes, metabolic disorders, hypertension and other vascular diseases. Recent studies showed that obesity-induced inflammation may be critical for IR. To investigate the anti-inflammatory effect of sodium butyrate (NaB) on obesity-induced inflammation, the db/db mice were intraperitoneally injected with NaB for 6 weeks. Glucose control wa...
متن کاملPuerarin Alleviates Neuropathic Pain by Inhibiting Neuroinflammation in Spinal Cord
Neuropathic pain responds poorly to drug treatments, and partial relief is achieved in only about half of the patients. Puerarin, the main constituent of Puerariae Lobatae Radix, has been used extensively in China to treat hypertension and tumor. The current study examined the effects of puerarin on neuropathic pain using two most commonly used animal models: chronic constriction injury (CCI) a...
متن کاملMorphine paradoxically prolongs neuropathic pain in rats by amplifying spinal NLRP3 inflammasome activation.
Opioid use for pain management has dramatically increased, with little assessment of potential pathophysiological consequences for the primary pain condition. Here, a short course of morphine, starting 10 d after injury in male rats, paradoxically and remarkably doubled the duration of chronic constriction injury (CCI)-allodynia, months after morphine ceased. No such effect of opioids on neurop...
