Development of <i>mPing</i> ?based activation tags for crop insertional mutagenesis

چکیده
Modern plant breeding increasingly relies on genomic information to guide crop improvement. Although some genes are characterized, additional tools needed effectively identify and characterize associated with traits. To address this need, the mPing element from rice was modified serve as an activation tag induce expression of nearby genes. Embedding promoter sequences in resulted a decrease overall transposition rate; however, effect negated by using hyperactive version called mmPing20. Transgenic soybean events carrying mPing-based tags appropriate transposase cassettes showed evidence transposition. Expression analysis line that contained heritable insertion mmPing20F indicated induced overexpression This represents significant advance gene discovery technology have potential more phenotypes than original element, improving effectiveness mutagenesis system.

T-DNA insertional mutagenesis for activation tagging in rice.
We have developed a new T-DNA vector, pGA2715, which can be used for promoter trapping and activation tagging of rice (Oryza sativa) genes. The binary vector contains the promoterless beta-glucuronidase (GUS) reporter gene next to the right border. In addition, the multimerized transcriptional enhancers from the cauliflower mosaic virus 35S promoter are located next to the left border. A total ...

editorial I n this issue of Molecular Therapy, David Rus-sell provides an invited commentary on data presented at the tenth annual meeting of the American Society of Gene Therapy that strongly suggested that adenovirus-associated virus (AAV) vector insertion could in certain models be linked to the development of cancer. 1 A short report describing the data that are the subject of this commenta...
متن کاملInsertional mutagenesis of pathogenic fungi.
Screening insertional mutants for loss of virulence is an effective method for investigating the molecular genetic basis of bacterial pathogenesis, but has only recently been applied to fungal pathogens. For many pathogenic fungi transformation with heterologous plasmid DNA results in complex integration events. This problem can now be circumvented for some species using restriction enzyme medi...

متن کاملTn7-based genome-wide random insertional mutagenesis of Candida glabrata.
We describe and characterize a method for insertional mutagenesis of the yeast pathogen Candida glabrata using the bacterial transposon Tn7. Tn7 was used to mutagenize a C. glabrata genomic fosmid library. Pools of random Tn7 insertions in individual fosmids were recovered by transformation into Escherichia coli. Subsequently, these were introduced by recombination into the C. glabrata genome. ...
