Development of a simple one-step multiplex RT-PCR system for simultaneous detection of DNA and RNA viruses of cucurbit leaf crumple virus, cucurbit yellow stunting disorder virus, squash vein yellowing virus, and cucurbit chlorotic yellows virus

چکیده
A highly sensitive one-step multiplex RT-PCR (OMRT-PCR) system with an internal plant gene control was developed to simultaneously detect single and mixed RNA DNA viral pathogens affecting watermelon squash in the Southeastern United States. Current methods rely on a singleplex detection for several viruses which is time-consuming large-scale field monitoring. The gene-specific primers one virus, cucurbit leaf crumple virus (CuLCrV), three viruses, yellow stunting disorder (CYSDV), vein yellowing (SqVYV), chlorotic yellows (CCYV), were used development of OMRT-PCR. gene, rubisco from cucurbits as standardization assay monitor false amplification. Surveys of yellowing viruses in plastic tunnels and in open field crops of melon (Cucumis melo cultivar catalupo), oriental melon (C. melo cultivar oriental melon), and cucumber (C. sativus) were carried out in two melon-growing areas in 2014, Korea. Severe yellowing symptoms on older leaves of melon and chlorotic spots on younger leaves of melon were observed in the plastic tunnels. The sympto...

Background: Cucurbit aphid-borne yellows virus (CABYV) is among the most important yellowing viruses on cucurbits in Iran. The presence of CABYV has been previously reported from the major cucurbit growing areas in Iran, however, there are few studies concerning the detection of the diff erent strains of this virus in the grower’s fi elds, and especially, there is no report of ...

Cucurbit chlorotic yellows virus (CCYV) (genus Crinivirus, family Closteroviridae) is an emerging plant virus, and is now spreading and causing severe economic losses to cucurbit crops in many Asian countries. CCYV is believed to be transmitted specifically by the sweetpotato whitefly, Bemisia tabaci, in a semipersistent manner. In the present study, we provide direct evidence for the semipersi...

An analysis of nucleotide sequences in five coding and one non-coding genomic regions of 35 Cucurbit yellow stunting disorder virus (CYSDV) isolates collected on a local scale over an 8 year period is reported here. In total, 2277 nt were sequenced for each isolate, representing about 13 % of the complete virus genome. Mean nucleotide diversity for the whole population in synonymous positions i...

Cucurbit yellow stunting disorder virus (CYSDV) is a destructive virus of cucurbits in Iran. During 2008-2012 growing seasons a total of 366 cucurbit samples including melon, cucumber, snakemelon and squash with typical symptoms of CYSDV infection were collected from ten Provinces in Iran. They were screened by ELISA and the infectivity of ELISA-positive samples was confirmed by RT-PCR. The res...
