Development of a microencapsulated probiotic containing Pediococcus acidilactici WU222001 against avian pathogenic Escherichia coli

Background and Aim: Probiotics are beneficial microorganisms for humans animals. In this study, we developed a microencapsulated probiotic with antibacterial activity against avian pathogenic Escherichia coli (APEC). Materials Methods: Alignment of the 16S rRNA sequences isolate WU222001 those deposited in GenBank revealed that was Pediococcus acidilactici 99.6% homology. This bacterium characterized as based on its tolerance toward vitro gastrointestinal tract (GIT) conditions, hydrophobicity, auto-aggregation. The probiotic’s culture supernatant APEC investigated using broth microdilution assay. sodium alginate agar diameters ranging from 47 to 61 µm. Then, physicochemical characteristics stability microcapsules were determined. Results: resistance low pH, bile salts, pancreatin, relative values 79.2%, 70.95%, 90.64%, respectively. Furthermore, exhibited 79.56% auto-aggregation 55.25% hydrophobicity at 24 h. strong clinical isolates minimum inhibitory concentration bactericidal 12.5% 25% v/v, Microencapsulation-enhanced bacterial viability GIT compared free cells. Moreover, 89.65% encapsulated cells released into simulated intestinal fluid within 4 viable count 63.19% after 3 months storage 4°C. Conclusion: results indicated P. inhibited growth APEC. addition, microencapsulation extends under harsh indicating potential application feed production.
