چکیده

Daphnia magna belongs to the Cladocera and plays an important role in water ecosystem. With intensification of pollution, wild population D. has declined rapidly recent years, insufficient molecular markers have limited effective research conservation this species. In research, 30 novel single nucleotide polymorphism (SNP) were developed a cultivar 12 using restriction site-associated DNA sequencing (RAD-seq). The minor allele frequency, observed heterozygosity, expected heterozygosity ranged from 0.115 0.483, 0.073 0.800, 0.077 0.520, respectively. PIC 0.071 0.403. Six loci showed significant deviations Hardy-Weinberg equilibrium after Bonferroni correction (p < 0.05).These newly polymorphic SNP for are great significance terms genetic breeding magna, identification artificially domesticated species genetics research.

Restriction Site-Associated DNA Sequencing (RAD-seq) Reveals an Extraordinary Number of Transitions among Gecko Sex-Determining Systems.

Sex chromosomes have evolved many times in animals and studying these replicate evolutionary "experiments" can help broaden our understanding of the general forces driving the origin and evolution of sex chromosomes. However this plan of study has been hindered by the inability to identify the sex chromosome systems in the large number of species with cryptic, homomorphic sex chromosomes. Restr...

Development of Transcriptomic Markers for Population Analysis Using Restriction Site Associated RNA Sequencing (RARseq)

We describe restriction site associated RNA sequencing (RARseq), an RNAseq-based genotype by sequencing (GBS) method. It includes the construction of RNAseq libraries from double stranded cDNA digested with selected restriction enzymes. To test this, we constructed six single- and six-dual-digested RARseq libraries from six F2 pitcher plant individuals and sequenced them on a half of a Miseq ru...

Small RNA Sequencing Based Identification of MiRNAs in Daphnia magna

Small RNA molecules are short, non-coding RNAs identified for their crucial role in post-transcriptional regulation. A well-studied example includes miRNAs (microRNAs) which have been identified in several model organisms including the freshwater flea and planktonic crustacean Daphnia. A model for epigenetic-based studies with an available genome database, the identification of miRNAs and their...

Assessing the phylogeographic history of the montane caddisfly Thremma gallicum using mitochondrial and restriction-site-associated DNA (RAD) markers

Repeated Quaternary glaciations have significantly shaped the present distribution and diversity of several European species in aquatic and terrestrial habitats. To study the phylogeography of freshwater invertebrates, patterns of intraspecific variation have been examined primarily using mitochondrial DNA markers that may yield results unrepresentative of the true species history. Here, popula...

Integrating restriction site-associated DNA sequencing (RAD-seq) with morphological cladistic analysis clarifies evolutionary relationships among major species groups of bee orchids

Background and Aims Bee orchids (Ophrys) have become the most popular model system for studying reproduction via insect-mediated pseudo-copulation and for exploring the consequent, putatively adaptive, evolutionary radiations. However, despite intensive past research, both the phylogenetic structure and species diversity within the genus remain highly contentious. Here, we integrate next-genera...
