Determination of Taurine in Quadriceps Femoris by High Performance Liquid Chromatography with Precolumn Derivatization

چکیده

A method for determination of taurine in mouse quadriceps femoris by high performance liquid chromatography with precolumn derivatization ophthalaldehyde (OPA) was established. The sample extracted and treated canon exchange resin. Results showed that separated quantified on C18 reversed phase column chromatographic (HPLC) after OPA 3min, using mixture methanol phosphate (V/V=1:1,pH=4.9) as mobile phase, rate flow is 0.6mL/min, detecting at 340nm UV-detector, L-Glutamine internal standard. result the linear ranger 6.25-187.7ng/mL, correlation coefficient R2=0.9994, recoveries were 91.8%-101.8%, RSD=3.2% (n=6). retention time 7.32 min. concentration 3.18mg/g. protein amino acid pre-treatment. good separation effect, simple, reliable, can be used to analyze mammal tissue.
