Detection of Mycovirus in Sclerotium Rolfsii, A Phytopathogenic Fungus in Bangladesh

A research project was conducted to detect presence of mycovirus, a probable virocontrol agent in Sclerotium rolfsii, destructive soil-borne plant pathogenic fungus. total 500 isolates the pathogen isolated from infected wheat, bush bean, tomato, eggplant, lentil, chickpea, chilli and okra grown Bangladesh following tissue planting methods brought Virology laboratory Okayama University, Japan. After Japan, 472 were re-isolated fungal cultured on potato dextrose agar for extraction DNA dsRNA. The extracted nucleic acids tested viruses using rolling circle amplification (RCA) (for viruses) cellulose column chromatography RNA viruses). detected S. rolfsii further characterized molecularly. For detection virus, 120 tested, none found positive viruses. dsRNA 7 showed possible band. partial cDNA sequences segments strain SR336 obtained. BlastX database search with sequence similarities randorna like virus. To screen virus isolates, 64 selected rolfsii. by RT-PCR specific primers. Out 13, (20.31%) rest absent virus.
