Detection of multiple mycoplasma infection in cell cultures by PCR.
A total of 301 cell cultures from 15 laboratories were monitored for mycoplasma (Mollicutes) using PCR and culture methodology. The infection was detected in the cell culture collection of 12 laboratories. PCR for Mollicutes detected these bacteria in 93 (30.9%) samples. Although the infection was confirmed by culture for 69 (22.9%) samples, PCR with generic primers did not detect the infection...
متن کاملDetection of mycoplasma contamination in cell cultures by a mycoplasma group-specific PCR.
The suitability of a 16S rRNA-based mycoplasma group-specific PCR for the detection of mycoplasma contamination in cell cultures was investigated. A total of 104 cell cultures were tested by using microbiological culture, DNA fluorochrome staining, DNA-rRNA hybridization, and PCR techniques. A comparison of the results obtained with these techniques revealed agreement for 95 cell cultures. Disc...
متن کاملMycoplasma Infection of Cell Cultures
This book is a collection of papers dealing with one of the most ubiquitous and troublesome nuisances of present day research in cell biology. Mycoplasma contamination of cell lines in a cell culture laboratory can cause changes in growth and biochemical properties of the cells, thus introducing uncontrolled variables into the experimental system. The papers in this book were presented at a wor...
متن کاملdetection of pestivirus contamination in cell cultures by nested-pcr
in this study a nested-pcr assay was optimized for detection of two bvdv biotype of nadl strain. a part of 5' non-coding region of virus, 249 bp in size, was amplified in rt-pcr. pcr product was cloned in a ptz57r/t vector and sequencing results confirmed the specificity of the test. internal primers were designed and a 155 bp dna fragment was amplified in nested-pcr. the sensitivity of rt...
متن کاملIsolation and detection of Mycoplasma pneumoniae from cell culture by culture and PCR.
Background: Mycoplasma pneumoniae is one of the major species of mycoplasmas which could contaminate cell cultures. Identification of M. pneuminae is significant because Mycoplasmas could contaminate and have unsuitable effect on the cell cultures. Rapid detection of these contaminations is so important and it could be significant role in preventing and controlling of contamination in cell cul...
