چکیده

Introduction: Water are important for human life. The Minister of Health the Republic Indonesia stipulates Number 492 2010 about drinking water quality requirements to ensure safe consumption. One free from Escherichia coli or total Coliform. When these bacteria found in water, it can cause diarrhea. In Province Central Java, diarrhea is biggest death children under five. Therefore, one locations Java was chosen, namely Jepara Regency, more precisely, District. Methods: This study used descriptive with a cross-sectional design. Data collection using sampling technique and obtained 28 samples. Then examination carried out at Regional Laboratory Regency filter membrane method Chromocult Coliform Agar Plate. Results: From samples, 11 samples (39.3%) were contaminated by 8 (72.72%) coli. Conclusion: Following up finding study, hoped that further researchers will carry researches on hygiene sanitation depot, so contaminations refill known.

Development of multiplex PCR for the detection of total coliform bacteria for Escherichia coli and Clostridium perfringens in drinking water.

Multiplex PCR amplification of lacZ, uidA and plc genes was developed for the simultaneous detection of total coliform bacteria for Escherichia coli and Clostridium perfringens, in drinking water. Detection by agarose gel electrophoresis yielded a band of 876 bp for the lacZ gene of all coliform bacteria; a band of 147 bp for the uidA gene and a band of 876 bp for the lacZ gene of all strains o...

Total coliform detection in drinking water: comparison of membrane filtration with Colilert and Coliquik.

The Colilert (CL) and Coliquik (CQ) systems were compared in a presence-absence format against the Standard Methods membrane filtration (MF) technique to determine whether differences existed in total coliform detection. Approximately 750 water samples were collected from distribution systems, covered and uncovered storage reservoirs, well sites, and the influent to drinking water treatment pla...

Detection of coliform organisms in drinking water by radiometric method.

The radiometric method has been used for detection of coliform bacteria in water. The method is based on measuring the released metabolic 14CO2 from 14C-lactose in growth media containing coliform organisms incubated at 37 degrees C under continuous shaking. This rapid and sensitive radiometric method permits the detection of even single coliform organisms within 6 hours of incubation. Using th...

Comparison of Escherichia coli, Total Coliform, and Fecal Coliform Populations as Indicators of Wastewater Treatment Efficiency

Escherichia coli, total coliform, and fecal coliform population data were collected from two wastewater treatment facilities, a subsurface flow artificial wetlands, and a receiving stream. Results are presented from individual wastewater treatment process streams, final effluent, and river sites upstream and downstream of the treatment [ acilities. The QuantiTray technique with 4-mcthylumbellif...

Comparing Defined- Substrate Coliform Tests for the Detection of Escherichia coli in Water

Two commercially available defined-substrate coliform tests were compared with EC medium plus 4-methylumbelliferyl-p-D-glucuronide (MUG) for detecting chlorine-exposed Escherichia coli in spiked water samples. Statistical analyses of the test results indicated no sign!nifcant differences in detection of E. cofi between the Autoanalysis Colilert test and EC medium with MUG. There were, however, ...
