Design of Beta-2 Microglobulin Adsorbent Protein Nanoparticles

Beta-2 microglobulin (B2M) is an immune system protein that found on the surface of all nucleated human cells. B2M naturally shed from cell surfaces into plasma, followed by renal excretion. In patients with impaired function, will accumulate in organs and tissues leading to significantly reduced life expectancy quality life. While current hemodialysis methods have been successful managing electrolyte as well small large molecule disturbances arising chronic failure, they shown only modest success plasma levels similar sized proteins, while sparing important proteins such albumin. We describe a systematic design effort aimed at adding ability selectively remove specific, undesired waste failure patients. A novel nanoparticle built using tetrahedral assembly scaffold presents 12 copies B2M-binding nanobody described. The designed binds specifically through protein–protein interactions nanomolar binding affinity (~4.2 nM). Notably, increases effective size over 50-fold, offering potential selective avenue for separation based size. present data support utility removing either size-based filtration or polyvalent stationary matrix under blood flow conditions.
