Deep Membrane Proteome Profiling of Rat Hippocampus in Simulated Complex Space Environment by SWATH

Despite the development and great progress in field of space biology, astronauts are still facing many challenges space. The environment which stay includes microgravity, noise, circadian rhythms disorder, confinement, has deep effect both on physiology psychology astronauts. It was reported that long-term flight could cause astronauts’ anxiety depression. However, underlying mechanism is not yet fully understood. Therefore, present study, rat tail suspension model with rhythms, confinement employed to simulate complex environment. We found rats exhibited depressive-like behavior by sucrose preference, forced swimming, open-field tests. membrane proteome hippocampus investigated “SWATH quantitation” technology control simulated (SCSE) groups. Out 4520 quantified proteins, 244 differentially expressed proteins were obtained between SCSE rats, functionally enriched a series biological processes, such as translation, protein phosphorylation, brain development, endocytosis, nervous system axonogenesis, vesicle-mediated transport. reduction level neurexin-2, light, medium, heavy polypeptide neurofilament, rab 18, synaptogyrin 1, syntaxin-1A an increase neuroligin-1, munc18, snapin, synaptotagmin XII, complexin-1, etc., may play key part Furthermore, GSK-3 ? upregulated mass spectrometry, further validated western blotting. results study do favor designing effective countermeasures for future spaceflight.
