چکیده
Abstract BACKGROUND Autophagy is a self-eating process that supports cancer cell survival. activation during drug treatment often prolongs cancerous survival, causing resistance, as seen in glioblastoma multiforme (GBM) to temozolomide (TMZ). Thus, blocking autophagy an appealing approach overcoming resistance. Long noncoding RNAs (LincRNAs) have emerged potential targets. LincRNA LINC00467 (renamed ARLINC) was recently identified inhibitor cells. However, ARLINC’s mechanism of inhibition and whether its modulation can suppress remains unknown. OBJECTIVE Research Question: Can restore the sensitivity cells chemotherapy? Hypotheses: 1) ARLINC regulates chemotherapy by inducing autophagy. 2) Overexpression increase sensitivity, measured viability assays METHODS Aim Endogenous levels GBM lines will be using qRT-PCR. Cell upregulated with pBabe-ARLINC (experiment) pBabe (control) then exposed increasing concentrations TMZ analyzed MTS assay. Student’s T-test used determine significance. 3) flux are transiently transfected compared empty vector via Cyto-ID RESULTS 1: Compared Normal Human Astrocytes (NHA), U87MG had higher levels, LN18 SF295 lower, LN229 around same ARLINC. 2: showed no significant decrease sensitivity. 3: flux. CONCLUSIONS/FUTURE WORK Results show effect upregulation on Future work involves knockdown experiments intracellular effects sensitization.

Long noncoding RNA MALAT1-regulated microRNA 506 modulates ovarian cancer growth by targeting iASPP
MALAT1, an important cancer-associated long noncoding RNA (lncRNA), contributes to the development and progression of several cancers. Disordered expression of MALAT1 has been observed in several cancers, including cervical cancer, breast cancer, and ovarian cancer. However, the exact effects and molecular mechanisms of MALAT1 in ovarian cancer progression are still unknown. Here, we investigat...
متن کاملThe novel long intergenic noncoding RNA UCC promotes colorectal cancer progression by sponging miR-143
The human genome contains thousands of long intergenic noncoding RNAs (lincRNAs). However, the functional roles of these transcripts and the mechanisms responsible for their deregulation in colorectal cancer (CRC) remain elusive. A novel lincRNA termed upregulated in CRC (UCC) was found to be highly expressed in human CRC tissues and cell lines. UCC levels correlated with lymph node metastasis,...
متن کاملSOX2OT, a long non-coding RNA involved in autophagy regulation
Summary: SOX2 overlapping transcript (SOX2OT) is a long non-coding RNA associated with cancer pathogenesis. It contributes to a variety of cellular functions and recent evidence propounds its association with autophagy process. It has been showed that SOX2OT can regulate the expression of different autophagy associated factors in human cells with different mechanisms, however more remains to ...
متن کاملVernalization-mediated epigenetic silencing by a long intronic noncoding RNA.
Vernalization is an environmentally-induced epigenetic switch in which winter cold triggers epigenetic silencing of floral repressors and thus provides competence to flower in spring. In Arabidopsis, winter cold triggers enrichment of tri-methylated histone H3 Lys(27) at chromatin of the floral repressor, FLOWERING LOCUS C (FLC), and results in epigenetically stable repression of FLC. This epig...
