Current understanding of CTLA-4: from mechanism to autoimmune diseases

چکیده
Autoimmune diseases (ADs) are characterized by the production of autoreactive lymphocytes, immune responses to self-antigens, and inflammation in related tissues organs. Cytotoxic T-lymphocyte antigen 4 (CTLA-4) is majorly expressed activated T cells works as a critical regulator inflammatory response. In this review, we first describe structure, expression, how signaling pathways CTLA-4 participate reducing effector T-cell activity enhancing immunomodulatory ability regulatory (Treg) reduce response, maintain homeostasis, autoimmune silence. We then focused on correlation between different ADs molecule regulates inhibits onset, progression, pathology various ADs. Finally, summarized current progress therapeutic target for

Cytotoxic T lymphocyte antigen-4 (CTLA-4) A49G polymorphism and autoimmune blood diseases.
OBJECTIVE The cytotoxic T lymphocyte associated antigen-4 (CTLA-4) is expressed on T lymphocytes, and inhibits the T-cell responses. In animal models, it has been shown that complete CTLA-4 deficiency was lethal due to massive infiltration of tissues by polyclonally proliferating lymphocytes. CTLA-4 A49G polymorphism, which has been suggested to reduce the inhibitory function of the CTLA-4 mole...

Immune system dysfunction is one of the key features in onset and development of cancer and autoimmunity. Cytotoxic T-lymphocyte-antigen-4 (CTLA-4), as a leader immune checkpoint plays a crucial effects in the regulation of immune suppression and tolerance. In this review, role of CTLA-4 and its three important polymorphisms (SNP), CTLA-4 +49A/G, CTLA-4 CT60 A/G and CTLA-4 −318C/T in developmen...

It can be classified etiologically into two groups: primary and secondary hypophysitis. Primary hypophysitis is, by definit...
متن کاملInfluence of CTLA-4 gene polymorphism in autoimmune and infectious diseases.
Cell-mediated immunity requires costimulatory activity to initiate or inhibit antigen-specific T-cell responses. CTLA-4 is an inhibitory receptor expressed by activated and regulatory T cells. The single nucleotide polymorphism (SNP) +49 A/G of the CTLA-4 gene alters intracellular distribution of CTLA-4, interleukin-2 production, and, as a consequence, T-cell proliferation. The aim of this stud...

CD152 or cytotoxic T lymphocyte antigen-4 (CTLA-4) is an essential receptor involved in the negative regulation of T cell activation. Because of its profound inhibitory role, CD152 has been considered a sound susceptible candidate in autoimmunity and a persuasive target for cancer immunotherapy for over a decade. However, the precise roles played by this molecule continue to emerge. In particul...
