Cryopreservation of coconut plumule using droplet vitrification

In the present investigation, four types of explants from mature zygotic embryos coconut, viz., whole upper cotyledonary region without haustorium, half plumule with a portion radicle and exclusively plumular tissue, were cultured in 12 different media combinations to find suitable explant which could be regenerated after cryopreservation. Explants pre-cultured medium 0.4 0.5 M sucrose for three days followed by dehydration PVS3 solution durations on sterile aluminum strip treating loading solution. Strips treated liquid nitrogen inside cryoflask until bubbling stopped quickly transferred cryovial stored minimum period 24 hours nitrogen. It was observed that alone or small outer tissue ideal fast vitro growth recovery plantlets coconut supplemented NAA alone. Addition glutamine (5 mg L-1), TDZ (1 L-1) (18 aided vigorous plantlets. control, survival rate ranged 60 90 per cent pre-grown containing various durations, whereas it 14 75 cryopreserved ones. Considering high (75%) regrowth (35%) study, there is much scope further improvement procedure right combination factors so as enhance complete injury plumules during cooling rewarming.
