Creative interior design by Plasmodium falciparum: Lipid metabolism and the parasite's secret chamber

چکیده
Malaria parasites conceal themselves within host erythrocytes and establish a necessary logistics system through the three-membrane layered structures of these cells. To this system, lipid metabolism is needed for de novo synthesis lipids recycling extracellular erythrocyte components. Cholesterol supply depends on its uptake from environment cytoplasm, but phospholipids can be synthesized their own. This differential production species creates unique modifications in profile parasitized erythrocytes, which turn may influence biophysical and/or mechanical properties organelles vesicles communication among them. Variations local membrane possibly transportation various molecules such as parasite-derived proteins, because efficiencies secretion, vesicle fusion budding are partly determined by profiles. Comprehensive understanding parasite's biophysics membranes provides fundamental knowledge about pathogenic organisms could lead to new anti-malarials.

Chloroquine-Resistant Haplotype Plasmodium falciparum Parasites, Haiti
Plasmodium falciparum parasites have been endemic to Haiti for >40 years without evidence of chloroquine (CQ) resistance. In 2006 and 2007, we obtained blood smears for rapid diagnostic tests (RDTs) and filter paper blots of blood from 821 persons by passive and active case detection. P. falciparum infections diagnosed for 79 persons by blood smear or RDT were confirmed by PCR for the small sub...

متن کاملGlutathione export from human erythrocytes and Plasmodium falciparum malaria parasites.
Glutathione export from uninfected human erythrocytes was compared with that from cells infected with the malaria parasite Plasmodium falciparum using two separate methods that distinguish between oxidized (GSSG) and reduced (GSH) glutathione. One involved enzymatic recycling with or without thiol-masking; the other involved rapid derivatization followed by HPLC. Glutathione efflux from uninfec...
متن کاملCentral carbon metabolism of Plasmodium parasites.
The central role of metabolic perturbation to the pathology of malaria, the promise of antimetabolites as antimalarial drugs and a basic scientific interest in understanding this fascinating example of highly divergent microbial metabolism has spurred a major and concerted research effort towards elucidating the metabolic network of the Plasmodium parasites. Central carbon metabolism, broadly c...
متن کاملEvidence for differences in erythrocyte surface receptors for the malarial parasites, Plasmodium falciparum and Plasmodium knowlesi
Human erythrocytes lacking various blood group determinants were susceptible to invasion by Plasmodium falciparum including Duffy-negative erythrocytes that are refractory to invasion by Plasmodium knowlesi. Erythrocytes treated with trypsin or neuraminidase had reduced susceptibility of P. falciparum and normal susceptibility to P. knowlesi. Chymotrypsin treatment (0.1 mg/ml) blocked invasion ...
