چکیده
Multicellular cancer spheroids (MCSs) have emerged as a promising in vitro model that recaptures many features of solid tumours vivo. To generate spheroids, cells are encapsulated microgels with high throughput. While the biophysical properties spheroid and biomaterial influence function behavior cells, characterization these remains largely unexplored. In addition, existing techniques lack control over cell positioning, resulting formation large variability. Herein, versatile, microfluidic platform for generating biocompatible core–shell uniform is reported. an situ micromechanics measuring device to determine stiffness individual artificial niches used. The power microfluidics-based method by making MCS-laden interactions stromal demonstrated. Together vivo data, it shown tumor proliferation promoted cocultured fibroblasts.

Ultrasoft, highly deformable microgels.
Microgels are colloidally stable, hydrogel microparticles that have previously been used in a range of (soft) material applications due to their tunable mechanical and chemical properties. Most commonly, thermo and pH-responsive poly(N-isopropylacrylamide) (pNIPAm) microgels can be fabricated by precipitation polymerization in the presence of the co-monomer acrylic acid (AAc). Traditionally pNI...
متن کاملMicrogels loaded with gold nanorods: photothermally triggered volume transitions under physiological conditions.
Photothermally driven volume transitions in polymer microgels have promising applications for site-specific drug delivery and photodynamic therapy. We studied the temperature-induced volume phase transitions for a series of thermoresponsive microgels of various compositions to find a system with a sharp transition in the physiologically relevant range spanning 38-41 degrees C in 0.01 M phosphat...
متن کاملDesign of biocompatible chitosan microgels for targeted pH-mediated intracellular release of cancer therapeutics.
We report the rational design of a chitosan-based drug delivery system. The chitosan derivative N-[(2-hydroxy-3-trimethylammonium)propyl]chitosan chloride (HTCC) was ionically cross-linked by sodium tripolyphosphate (TPP) to form sub-200-nm microgels that are responsive to pH changes. When these microgels were loaded with methotrexate disodium (MTX), a cytotoxic drug for cancer treatment, and c...
متن کاملConstructing 3D heterogeneous hydrogels from electrically manipulated prepolymer droplets and crosslinked microgels
Formation of multifunctional, heterogeneous, and encoded hydrogel building blocks, or microgels, by crosslinking and assembly of microgels are two essential steps in establishing hierarchical, complicated, and three-dimensional (3D) hydrogel architectures that recapitulate natural and biological structures or originate new materials by design. However, for the variety of the hydrogel materials ...
متن کاملNotch Effects Under Physiologically-Relevant Conditions of Conventional and Highly Crosslinked UHMWPEs
Introduction: Ultra high molecular weight polyethylene (UHMWPE) components used in total joint replacements have rims and fillets that act as design stress risers; thus, it is of interest to determine the behavior UHMWPE under multiaxial loading conditions. Currently, there are a number of conventional and crosslinked UHMWPE formulations in clinical use in total hip and total knee replacement c...
