Construction of DNA ladder for determination of small size DNA fragments

DNA marker is commonly used to determine the size of fragments by electrophoresis in routine molecular biology laboratories. In this study, we report a new procedure prepare recombinant plasmids pSY-60 which was partially digested one restriction enzyme for generating markers 7 from 60 420 bp. The included synthesis bp fragment with EcoRI sites at both ends using PCR extension, self-ligation and subcloning ligated product into plasmid, pSY-60. Once being cloned, 500 ng purified 100 µL incompletely EcoRI, sufficiently producing 50 acrylamide gels. Our production could be simple, time saving inexpensive comparison current ones widely most
