Connecting Hydrophobic Surfaces in Cyclic Peptides Increases Membrane Permeability

نویسندگان

چکیده

Abstract N‐ or C‐methylation in natural and synthetic cyclic peptides can increase membrane permeability, but it remains unclear why this happens some cases not others. Here we compare three‐dimensional structures for from six families, including isomers differing only the location of an Cα‐methyl substituent. We show that a single methyl group increases permeability when connects expands hydrophobic surface patches. Positional isomers, with same molecular weight, hydrogen bond donors/acceptors, rotatable bonds, calculated LogP, topological polar area, total have different permeabilities correlate size largest continuous patch. These results illuminate key local determinant permeability.

Hydrophobic interactions of peptides with membrane interfaces.

The thermodynamic principles underlying the structural stability of membrane proteins are difficult to obtain directly from whole proteins because of intractable problems related to insolubility in the aqueous phase and extreme stability in the membrane phase. The principles must therefore be surmised from studies of the interactions of small peptides with lipid bilayers. This review is concern...

Membrane permeability of hydrocarbon-cross-linked peptides.

Schafmeister, Po, and Verdine (another study) introduced a method using a hydrocarbon linker (staple) to stabilize a peptide in a helical configuration. One intended goal of this scheme is to facilitate the delivery of peptide drugs into target cells. Here, we investigate whether stapled peptides are intrinsically membrane permeable, by performing a case study on a stapled 12-mer peptide named ...

Eosinophil peroxidase increases membrane permeability in mammalian urinary bladder epithelium.

Eosinophil peroxidase (EPO), a cationic protein found in eosinophils, has been reported to be cytotoxic independent of its peroxidase activity. This study investigated with electrophysiological methods whether EPO is toxic to mammalian urinary bladder epithelium. Results indicate that EPO, when added to the mucosal solution, increases apical membrane conductance of urinary bladder epithelium on...

Conformational flexibility, internal hydrogen bonding, and passive membrane permeability: successful in silico prediction of the relative permeabilities of cyclic peptides.

We report an atomistic physical model for the passive membrane permeability of cyclic peptides. The computational modeling was performed in advance of the experiments and did not involve the use of "training data". The model explicitly treats the conformational flexibility of the peptides by extensive conformational sampling in low (membrane) and high (water) dielectric environments. The passiv...

Functional implications of modifying RyR-activating peptides for membrane permeability.

1. Our aim was to determine whether lipoamino acid conjugation of peptides that are high-affinity activators of ryanodine receptor (RyR) channels would (a) render the peptides membrane permeable, (b) alter their structure or (a) reduce their activity. The peptides correspond to the A region of the II-III loop of the skeletal dihydropyridine receptor. 2. The lipoamino acid conjugation increased ...
