چکیده
Objective To compare specificity and sensitivity of a commercially available fixed cell-based assay (F-CBA) to radioimmunoprecipitation (RIPA) for acetylcholine receptor antibody (anti-AChR) detection in myasthenia gravis (MG). Background Approximately 50% ocular 85% generalized MG are anti-AChR positive by RIPA, the current gold standard test. Clustered live (L-CBA) can detect low-affinity that missed but costly time-consuming nature L-CBA has restricted its use specialized centres. A commercial F-CBA become detection, diagnostic performance compared RIPA requires evaluation. Design/Methods In this retrospective cohort study we reviewed clinical information suspected patients evaluated at London Health Sciences Centre clinic, who were clinically classified as or non-MG had then performed. Classification each patient F-CBA-negative/positive, RIPA-negative/positive, MG/non-MG permitted calculations assay. Results Six-hundred-eighteen included analysis. The median age time sample collection was 45.8 years (range: 7.5–87.5 years) 312/618 (50.5%) female. Of 618 patients, 395 (63.9%) MG. Specificity both excellent (99.6% vs. 100%, P > 0.99). One F-CBA-positive non-MG, although retrospect with functional overlay challenging exclude. Sensitivity significantly higher than (76.7% 72.7%, = 0.002). Overall, 20/97 (21%) otherwise SNMG after evaluation detected F-CBA. Conclusions our specificity, while 4% 21% patients. Our findings indicate is viable alternative detection.

TE671 cell-based ELISA for anti-acetylcholine receptor antibody determination in myasthenia gravis.
BACKGROUND Acetylcholine receptor (AChR) from human muscles is the antigen used currently in radioimmunoprecipitation assays (RIPAs) for the determination of anti-AChR antibodies in the diagnosis of myasthenia gravis (MG). Our aim was to develop and validate an ELISA using TE671 cells as the source of AChR. METHODS After TE671 cell homogenization, the crude AChR extract was used for plate coa...

In a retrospective case note study of 86 patients with myasthenia gravis, 60 had an anti-acetylcholine receptor antibody assay performed by the regional immunology laboratory. Antibody was detected in 38% which compares with 66-93% in other series. Whilst the use of staphylococcal protein A to precipitate the antibody-receptor complex, rather than anti-human immunoglobulin, may be partly respon...

BACKGROUND: Aflatoxin B1 (AFB1) is a toxic metaboliteproduced by Aspergillus species that contaminates a wide range ofagricultural products. OBJECTIVES: This study was designed todevelop a rapid and highly sensitive immunoassay method inmicroarray format for quantitative detection of AFB1 to evaluatethe potential of microarray platform for high-throughput screening, which can be beneficial in fo...
متن کاملDevelopment of a recombinant protein-based dot-blot hybridization assay for the detection of antibody to chicken infectious bronchitis virus
Nucleocapsid (N) protein of infectious bronchitis virus (IBV), one of the viral structural proteins, inducesstrong antibody response in natural infection. In this study, a simple, recombinant N protein-based dot-blottest was developed to serologically examine chicken serum samples for the presence of IBV antibody.Initially, 72 serum samples were tested for the presence of IBV antibody using a c...
