Combining collagen extraction with mineral Zn isotope analyses from a single sample for robust palaeoecological investigations

چکیده

Abstract Collagen extraction from bones or dentine, commonly used for radiocarbon ( 14 C) dating and stable carbon nitrogen isotope (δ 13 C δ 15 N) analyses, involves the dissolution of bioapatite skeletal elements. This fraction is typically disposed during pretreatment. Here, we test possibility utilising this dissolved mineral solution analysis zinc composition 66 Zn). Bioapatite Zn a novel trophic level indicator similar to collagen N but with isotopic fractionation independent nitrogen, thus providing additional dietary information. We tested ways minimise contamination phase extraction. then archaeological bone samples Ain Difla (Jordan) Ranis (Germany) compare values following our protocol same sample material in metal-free cleanroom. Our results demonstrate that only minor adjustments contamination, protocols employed (palaeo)dietary can be analyses even when does not take place allows us gain an proxy complement interpretations perform more robust (palaeo)ecological investigations without further destructive sampling.

New sample holder geometry for high precision isotope analyses

Secondary ion mass spectrometry is applied to a wide range of Geoscience applications because of its capability to provide direct in situ measurement of elemental and isotopic composition. The CAMECA IMS 1280 and 1280-HR are large geometry ultra-high sensitivity ion microprobes that provide excellent precision and reproducibility for isotope ratio measurements. A precision at the tenth permil l...

MPLEx: a Robust and Universal Protocol for Single-Sample Integrative Proteomic, Metabolomic, and Lipidomic Analyses

Integrative multi-omics analyses can empower more effective investigation and complete understanding of complex biological systems. Despite recent advances in a range of omics analyses, multi-omic measurements of the same sample are still challenging and current methods have not been well evaluated in terms of reproducibility and broad applicability. Here we adapted a solvent-based method, wide...

Tunable Single-Cell Extraction for Molecular Analyses

Because of cellular heterogeneity, the analysis of endogenous molecules from single cells is of significant interest and has major implications. While micromanipulation or cell sorting followed by cell lysis is already used for subsequent molecular examinations, approaches to directly extract the content of living cells remain a challenging but promising alternative to achieving non-destructive...

Combining stable isotope analyses and geolocation to reveal kittiwake migration

Determining migratory strategies of seabirds is still a major challenge due to their relative inaccessibility. Small geolocators are improving this knowledge, but not all birds can be tracked. Stable isotope ratios in feathers can help us to understand migration, but we still have insufficient baseline knowledge for linking feather signatures to movements amongst distinct water masses. To under...

Simplifying and improving the extraction of nitrate from freshwater for stable isotope analyses.

Determining the isotopic composition of nitrate (NO(3)(-)) in water can prove useful to identify NO(3)(-) sources and to understand its dynamics in aquatic systems. Among the procedures available, the 'ion-exchange resin method' involves extracting NO(3)(-) from freshwater and converting it into solid silver nitrate (AgNO(3)), which is then analysed for (15)N/(14)N and (18)O/(16)O ratios. This ...
