Abstract Background Gloriosa superba L. belongs to Colchicaceae, which is an important medicinal plant containing high-priced alkaloid colchicines and other potent phytochemicals. Due its extensive importance at the industrial level, this overexploited. Moreover, indiscriminate harvesting for raw material leads a decline in population of natural environment. Thus, present study deals with optimization colchicine accumulation from different intervals vitro vivo tubers . Result To obtain tuberization, shoot tip explants were inoculated on Murashige Skoog medium prepared 3 mg/l BA 0.5 Kn + 1 2, 4-D followed by 2 0.2 NAA. In high-performance thin-layer chromatography (HPTLC), linearity range was set concentration 100–1000 ng/spot regression value (r) 0.99. Its R f (0.25) recorded 254 nm. The amount tuber 7.75 ± 0.25% dry weight, while nearby 7.7 0.40% weight produced weeks old tuber. This 4th-week 6.35 0.17% then gradual decrease accumulation. Conclusion significant results stages observed. The clinical features of colchicine toxicity in a patient following ingestion of Gloriosa superba tubers are described. Gastroenteritis, acute renal failure, cardiotoxicity and haematological abnormalities were the main toxic manifestations. There was no hypotension and no neurological manifestations.
