چکیده

ObjectiveThe aim of this study was to assess the clinical utility metagenomic next-generation sequencing (mNGS) on smear-negative extrapulmonary specimens collected in China.MethodsSpecimens were tested by mNGS and other routine tests for tuberculosis (TB). The diagnostic accuracy calculated compared with final diagnosis.ResultsThe sensitivity found be significantly higher than sensitivities TB tests. Receiver operating characteristic curve analysis showed that achieved highest area under (AUC) value 0.79. positive rate tuberculous meningitis. All non-tuberculous pathogens directly simultaneously detected.ConclusionsmNGS appeared superior all previous etiological could identify possible at once within 48 h.

RAPID DETECTION OF MYCOBACTERIUM TUBERCULOSIS IN CLINICAL SPECIMENS BY POLYMERASE CHAIN REACTION

We investigated the use of DNA amplification by polymerase chain reaction (peR) for detection of Mycobacterium tuberculosis in 300 patients who were suspected of having pulmonary tuberculosis and compared the results with culture results which were performed in parallel with PCR. Two-thirds of each sample was processed for smear and culture by standard methods and one-third was prepared fo...

Next-Generation Sequencing of Mycobacterium tuberculosis.

Technical Appendix Figure. Whole-genome, short-sequencing reads of Mycobacterium tuberculosis strain 4542 mapped to complete genome of reference strain H37Rv (NC_00962.3) by using Geneious 9.0 package: genome locus of spacers 7–9 in DR/CRISPR locus. Spacer numbers are given according to 43-spoligotyping (1). DR/CRISPR locus in M. tuberculosis is located on complementary strand, whereas spacers ...

Next-generation sequencing for identifying pyrazinamide resistance in Mycobacterium tuberculosis.

TO THE EDITOR—We read with interest the recent article by Kurbatova et al [1]. Pyrazinamide (PZA) is a key component of multidrug antituberculosis therapy, in both firstand second-line regimens. In their article, Kurbatova and colleagues underscore the public health importance of describing the epidemiology of PZAresistantMycobacterium tuberculosis (MTB) infection in the United States. Resistan...

Frequency of Smear-Negative Tuberculosis in Northwest Iran

Background: Microscopic smear examination is the most common test in tuberculosis (TB) detection. It is, however, not strong enough to identify TB in the majority of afflicted individuals; thus, a significant number of TB patients are smear negative and capable of transmitting the infection. The aim of this study was to evaluate the rate of smear-negative TB in northwest Iran. Methods: In this c...
