چکیده

Two Streptomyces strains, named N11-26 and DC10-5, were isolated from deep-sea non-photosynthetic stony coral, respectively. Strain produces lobophorin C divergolides, which are antimicrobial substances. This study aimed to classify these strains reveal their cryptic potential synthesize other secondary metabolites, such as polyketides nonribosomal peptides. Strains DC10-5 showed 16S rRNA gene sequence similarities of 100% 99.9% olivaceus NRRL B-3009T, By digital DNA–DNA hybridization using whole-genome sequences, classified olivaceus. was closer the type strain S. than possessed 17 clusters polyketide synthase (PKS) and/or peptide synthetases (NRPS) genes, whereas harbored 19 clusters. Putative products by predicted bioinformatic analyses. Although 15 conserved between two four specific in represents a diversity compounds To best our knowledge, this is first report annotating all PKS NRPS with putative provide useful information for genome mining.

This article introduces DoBISCUIT (Database of BIoSynthesis clusters CUrated and InTegrated, http://www.bio.nite.go.jp/pks/), a literature-based, manually curated database of gene clusters for secondary metabolite biosynthesis. Bacterial secondary metabolites often show pharmacologically important activities and can serve as lead compounds and/or candidates for drug development. Biosynthesis of...

Wide Distribution of Foxicin Biosynthetic Gene Clusters in Streptomyces Strains – An Unusual Secondary Metabolite with Various Properties

Streptomyces diastatochromogenes Tü6028 is known to produce the polyketide antibiotic polyketomycin. The deletion of the pokOIV oxygenase gene led to a non-polyketomycin-producing mutant. Instead, novel compounds were produced by the mutant, which have not been detected before in the wild type strain. Four different compounds were identified and named foxicins A-D. Foxicin A was isolated and it...

Diverse and Abundant Secondary Metabolism Biosynthetic Gene Clusters in the Genomes of Marine Sponge Derived Streptomyces spp. Isolates

The genus Streptomyces produces secondary metabolic compounds that are rich in biological activity. Many of these compounds are genetically encoded by large secondary metabolism biosynthetic gene clusters (smBGCs) such as polyketide synthases (PKS) and non-ribosomal peptide synthetases (NRPS) which are modular and can be highly repetitive. Due to the repeats, these gene clusters can be difficul...

Recent development of antiSMASH and other computational approaches to mine secondary metabolite biosynthetic gene clusters.

Many drugs are derived from small molecules produced by microorganisms and plants, so-called natural products. Natural products have diverse chemical structures, but the biosynthetic pathways producing those compounds are often organized as biosynthetic gene clusters (BGCs) and follow a highly conserved biosynthetic logic. This allows for the identification of core biosynthetic enzymes using ge...
