Citrinin Derivatives From Penicillium Citrinum Y34 That Inhibit ?-Glucosidase and ATP-Citrate Lyase

Two new citrinin dimers bearing a 6,6-spiroketal moiety (1 and 2) four known analogues (3–6), together with 18 monomers (7–24), were isolated from the culture of hydrothermal vent-associated fungus Penicillium citrinum Y34. Their structures identified by extensive spectroscopic analyses, 13 C NMR calculation in combination DP4+, linear correlation coefficient ( R 2 ), mean absolute error (MAE) values electronic circular dichroism (ECD) calculation. The ? -glucosidase ATP-citrate lyase (ACL) inhibitory activities compounds evaluated. Compounds 1, 3, 12 displayed moderate IC 50 239.8, 176.2, 424.4 ?M, respectively. Enzyme kinetics investigations 1 3 suggested their non-competitive inhibition K i 204.3 212.7 Meanwhile, compound 4 showed significant ACL potential an value 17.4 ?M. Furthermore, interactions investigated molecular docking assay. This study demonstrates that citrinins, especially for dimers, could be lead development agents treatment metabolic diseases.
