چکیده
Purpose: To investigate the effects of circ_0001953/miR-186 on human retinal vascular endothelial cell (HRVEC) injury evoked by high glucose. Methods: A model (HG group) was established using glucose-treated HRVECs, while untreated HRVECs were used as control group (Con group). The levels endothelin-1 (ET-1), ICAM-1 and IL-6 evaluated ELISA content malondialdehyde (MDA) superoxide dismutase (SOD) in determined. Apoptosis rate tested adopting flow cytometry. interrelationship between circ_0001953 miR-186 assessed dual luciferase reporter assay. Measurement Bax Bcl-2 implemented via western blot. Results: In HG group, increased downregulated, ET-1, IL-6, ICAM-1, apoptosis accompanied with up-regulated declined protein level. Furthermore, MDA SOD decreased. MiR-186 a target circ_0001953. Conclusion: Inhibition can repress inflammation, oxidative stress up-regulating expression miR-186.

Inhibition of miR-186-5p contributes to high glucose-induced injury in AC16 cardiomyocytes
A growing body of evidence has demonstrated that microRNAs (miRs) have pivotal roles in the pathophysiological development mechanisms of diabetic cardiomyopathy (DCM). Previous studies have demonstrated that miR-186-5p was significantly decreased in DCM. In addition, it has recently been reported that an imbalance of miR-186 is associated with a variety of physiological and pathological process...
متن کاملHigh glucose concentration induces endothelial cell proliferation by regulating cyclin‐D2‐related miR‐98
Cyclin D2 is involved in the pathology of vascular complications of type 2 diabetes mellitus (T2DM). This study investigated the role of cyclin-D2-regulated miRNAs in endothelial cell proliferation of T2DM. Results showed that higher glucose concentration (4.5 g/l) significantly promoted the proliferation of rat aortic endothelial cells (RAOECs), and significantly increased the expression of cy...

The present study investigated the role of echinocystic acid (EA) on the expression of nuclear factor (NF)-κB and cytochrome P450 1A1 (CYP1A1), and aortic morphology, in a rat model of hyperhomocysteinemia (Hhcy). A total of 50 Sprague Dawley rats were randomly divided into five groups as follows: Normal control (NC), model control (MC), vitamin control (VC; folic acid 1 mg/kg + vitamin B2 2 mg...
متن کاملTaurine prevents high-glucose-induced human vascular endothelial cell apoptosis.
Elevated blood glucose in uncontrolled diabetes is causally correlated with diabetic microangiopathy. Hyperglycemia-triggered accelerated endothelial cell apoptosis is a critical event in the process of diabetes-associated microvascular disease. The conditionally semiessential amino acid taurine has been previously shown to protect against human endothelial cell apoptosis. Therefore, this study...
متن کاملEffect of miR-200b on retinal endothelial cell function under high glucose environment.
As one of the important complications of diabetes, diabetic retinopathy (DR) presented high incidence worldwide. Hyperglycemia is an important promoting factor for DR occurrence and development. It can damage retinal endothelial cell, resulting in retinal structure and function disorder. Studies have shown that miR-200b may involve in regulating DR occurrence and development, but its specific f...
