چکیده
Introduction Ovarian cancer (OC) is the malignant tumor with highest mortality among gynecological cancers. Chemotherapy resistance a major obstacle to OC therapy. Circular RNAs (circRNAs) play crucial roles in development and chemoresistance. However, role potential mechanism of has-circ-001567 (circ-VPS13C) chemoresistance remain unknown. Material methods The levels circ-VPS13C, miR-106b-5p 14-3-3 zeta (YWHAZ) were detected by quantitative real-time polymerase chain reaction (qRT-PCR) or western blot assay. Cell Counting Kit-8 (CCK-8) assay was used assess cell viability calculate half inhibition concentration (IC50) cisplatin (DDP). autophagy-related markers measured apoptosis migration evaluated flow cytometry transwell assay, respectively. binding relationship between circ-VPS13C YWHAZ confirmed dual-luciferase reporter Xenograft performed explore vivo. Results Circ-VPS13C up-regulated, while down-regulated DDP-resistant tissues cells. Knockdown enhanced DDP sensitivity repressing autophagy increased via sponging miR-106b-5p. Moreover, directly targeted YWHAZ. Up-regulation alleviated decrease caused depletion. In addition, silencing decreased Conclusions knockdown through modulation miR-106b-5p/YWHAZ axis, providing new biomarker for improving efficacy chemotherapy.

miR-93/miR-106b/miR-375-CIC-CRABP1: a novel regulatory axis in prostate cancer progression.
Capicua (CIC) has been implicated in pathogenesis of spinocerebellar ataxia type-1 (SCA1) neurodegenerative disease and some types of cancer; however, the role of CIC in prostate cancer remains unknown. Here we show that CIC suppresses prostate cancer progression. CIC expression was markedly decreased in human prostatic carcinoma. CIC overexpression suppressed prostate cancer cell proliferation...
متن کاملLncRNA NEAT1 enhances the radio-resistance of cervical cancer via miR-193b-3p/CCND1 axis
LncRNAs have become a hot topic in various cancer-related researches. Radio-resistance is a great threat for cancer therapy. However, how lncRNAs affect the radio-resistance in cervical cancer is masked. As for our paper, it was discovered that NEAT1 was highly expressed in cervical cancer tissues and non-sensitive tissues as well as radio-resistant cell lines. And the overexpression of NEAT1 a...
متن کاملMALAT1/miR-101-3p/MCL1 axis mediates cisplatin resistance in lung cancer
In this study, we investigated the mechanism by which lncRNA metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) mediates cisplatin resistance in lung cancer. Lung cancer patients with high MALAT1 levels were associated with cisplatin resistance and low overall survival. Moreover, cisplatin-resistant A549/DDP cells showed higher MALAT1 expression than cisplatin-sensitive lung cancer...
متن کاملBerberine sensitizes ovarian cancer cells to cisplatin through miR-21/PDCD4 axis.
Recent studies have shown that microRNA-21 (miR-21) contributes to tumor resistance to chemotherapy. Interestingly, we have found that berberine could inhibit miR-21 expression in several cancer cell lines. In this study, we investigated whether berberine could modulate the sensitivity of ovarian cancer cells to cisplatin and explored the mechanism. The cisplatin-resistant SKOV3 cells that were...
متن کاملmiR-199a-3p enhances cisplatin sensitivity of ovarian cancer cells by targeting ITGB8
Drug resistance remains a large obstacle for the treatment of ovarian cancer. miRNAs have been reported to be involved in cisplatin (CDDP) resistance in ovarian cancer. The aim of the present study was to investigate the function and mechanism of miR-199a-3p in the CDDP resistance in ovarian cancer. We found that miR-199a-3p was significantly downregulated in chemoresistant ovarian cancer tissu...
