چکیده
This study was aimed to differentiate parental genomes, examining intergenomic composition, and mapping mitotic metaphase chromosomes by localizing 18S rDNA probes in seven interspecific hybrid progenies that originated from Lilium longiflorum. Since single method of situ hybridization has been widely used lily breeding, while this study, flow cytometry conjunction with genomic fluorescent determine the contribution each parent progenies. A significant variation observed DNA content, chromosome length, loci F1 as compared female male parents. L. longiflorum showed nearly two times higher content than parents × Asiatic progenies, but eight hansonii. Genomic results revealed both contributed an equal number their offspring. Fluorescent had 8, 6 7 parents, i.e., White heaven, Bright tower, respectively, whereas cultivar hansonii 8 12 respectively. Interspecific LA, 10–11 LM hybrids. These cytogenetic implied genetic chromosomal Therefore, combined potential be genome specific time-saving approach breeding could status hybrids origin achieving physical detecting genes different genomes.

Physical mapping of 18S-25S rDNA and 5S rDNA in Lupinus via fluorescent in situ hybridization.
Double-target fluorescent in situ hybridization (FISH) was used to determine the genomic distribution of ribosomal RNA genes in five Lupinus species: L. cosentinii (2n=32), L. pilosus (2n=42), L. angustifolius (2n=40), L. luteus (2n=52) and L. mutabilis (2n=48). 18S-25S rDNA and 5S rDNA were used as probes. Some interspecific variation was observed in the number and size of the 18S-25S rDNA loc...

The genomic in situ hybridization (GISH) technique was applied to Passiflora interspecific F1 HD13-133 hybrids (Passiflora sublanceolata x Passiflora foetida) and HD15-101 (Passiflora gardineri x Passiflora gibertii), and the backcrossed hybrids (BC1) HD18-106 and HD18-113 (Passiflora sublanceolata x HD13-133). GISH was performed using genomic probes prepared with the DNA from the paternal geni...
متن کاملMapping chromosome rearrangement breakpoints to the physical map of Caenorhabditis elegans by fluorescent in situ hybridization.
A scheme for rapidly mapping chromosome rearrangements relative to the physical map of Caenorhabditis elegans is described that is based on hybridization patterns of cloned DNA on meiotic nuclei, as visualized by fluorescent in situ hybridization. From the nearly complete physical map, DNA clones, in yeast artificial chromosomes (YACs), spanning the rearrangement breakpoint were selected. The p...

Half-smooth tongue sole (Cynoglossus semilaevis) is an important aquaculture flatfish in China. Cytogenetic analysis has revealed that its sex determination system is female heterogametic (ZZ/ZW). The W chromosome is morphologically larger and has been considered evolutionarily younger than any other chromosome in the set. However, the genetic origin and evolution process of this neo-chromosome...

In situ hybridization of 18S-5.8S-25S rDNA probes labeled with biotin or rhodamine and 5S rDNA probes labeled with digoxigenin was used to locate rDNA sites on root-tip metaphase chromosomes of Citrus sinensis L. (2n = 2x = 18), Poncirus trifoliata L. Raf. (2n = 2x = 18), and Citrus x Poncirus hybrids (2n = 2x = 18). Counterstaining with the fluorochromes chromomycin A3 and DAPI uniquely identi...
