Characterization of Adipose‐derived Stem Cells from Breast Cancer Cell Transplanted Mice

چکیده

Mesenchymal stem cells (MCSs) from adipose tissue are reported to have a pronounced impact on tumor growth or suppression. However, so far, how the cancer affect in vivo conditions has not been clarified. In this study, we evaluated of breast characteristics adipose-derived (ADSCs) under conditions. ADSCs were obtained Balb/c mice three consecutive weeks after transplantation with 1×106 MCF7 cells. The results showed that amount per one gram fat reduced over time, but reduction was significantly different. fact, isolated presenting specific MSC surface markers increased at second passage, compared time isolation. Particularly, they highly positive for CD90 (a indicator) and negative CD45 hematopoietic marker). Moreover, vitro culture, successfully differentiated into adipocytes, which detected by Oil Red O staining. This present experiment gives initial assessments ADSC characteristics; however, further investigations necessary fully evaluate differences cell potency immunophenotype caused transplantation.

Induction of T Regulatory Subsets from Naïve CD4+ T Cells after Exposure to Breast Cancer Adipose Derived Stem Cells

Background: Adipose derived stem cells (ASCs) provoke the accumulation and expansion of regulatory T cells, leading to the modulation of immune responses in tumor microenvironment. Objective: To assess the effect of tumoral ASCs on the trend of regulatory T cells differentiation. Methods: Peripheral blood naïve CD4+ T cells were co-cultured with ASCs derived from breast cancer or normal breast ...

Evaluation of Stem Cell Markers, CD44/CD24 in Breast Cancer Cell Lines

Background Cancer stem cells play crucial roles in resistance to therapeutic schemes and relapse of disease, so it is important to find targeted therapies that kill them selectively. Breast cancer is the most common cancer in females living in all part of the world including Iran and it has an important burden in public health with direct impact on patients’ families. Breast cancer in young ad...

Breast cancer stem cells.

Breast cancer stem cells (BCSCs) constitute a subpopulation of tumor cells that express stem cell-associated markers and have a high capacity for tumor generation in vivo. Identification of BCSCs from tumor samples or breast cancer cell lines has been based mainly on CD44(+)/CD24(-/low) or ALDH(+) phenotypes. BCSCs isolation has allowed the analysis of the molecular mechanisms involved in their...

Breast Cancer Stem Cells

Over 150 years ago, Cohnheim and Durante formalized the concept that cancers might arise from a small subset of cells with stem cell properties 1-3, and in 1961, Till and McCulloch demonstrated for the first time that the existence of hematopoietic stem cells (HSC) in the bone marrow, which was postulated that stem-like cells might be the origin of cancer 4. However, only recently did an increa...

Breast cancer stem cells

Cancer metastasis, resistance to therapies and disease recurrence are significant hurdles to successful treatment of breast cancer. Identifying mechanisms by which cancer spreads, survives treatment regimes and regenerates more aggressive tumors are critical to improving patient survival. Substantial evidence gathered over the last 10 years suggests that breast cancer progression and recurrence...
