Cerebrospinal fluid concentrations of hypoxanthine, xanthine, uridine and inosine: high concentrations of the ATP metabolite, hypoxanthine, after hypoxia.

CSF obtained for clinical purposes from newborn, children and adults has been analysed by high pressure liquid chromatography for hypoxanthine, xanthine, inosine, uridine and urate. Large rises in hypoxanthine and to a lesser extent xanthine occur for about 24 h after hypoxia. High concentrations were associated with later evidence of brain damage or subsequent death. Changes in CSF could be in...

Simultaneous liquid-chromatographic determination of hypoxanthine, xanthine, urate, and creatinine in cerebrospinal fluid, with direct injection.

In this method for simultaneously determining hypoxanthine, xanthine, urate, and creatinine in cerebrospinal fluid, centrifuged sample is directly injected on a reversed-phase liquid-chromatographic column. On elution with potassium phosphate buffer the compounds are quantified by their absorbance at 260 nm. Random error (CV) was between 1.2 and 3.4% and analytical recoveries were 99-104% at ph...

Enhanced ATP and GTP synthesis from hypoxanthine or inosine after myocardial ischemia.

Increasing therapeutic use is made of purines for the treatment of ischemic heart disease, but little is known about regulatory mechanisms involved. Therefore we perfused isolated rat hearts with 0.02 mmol/l [8-14C]hypoxanthine or inosine. Under normoxic conditions about 1% is taken up by the heart and partially used for synthesis of ATP and GTP at a rate of 0.4 and 0.1 nmol X min-1 X g dry wt-...

Liquid-chromatographic determination of inosine, xanthine, and hypoxanthine in uremic patients receiving hemodialysis treatment.

We used a liquid-chromatographic method to measure xanthine, hypoxanthine, and inosine in plasma of uremic patients before and after hemodialysis. The concentrations of all three were higher than in normal subjects. After dialysis treatment, the values were significantly lower than before dialysis, but still above normal.

Adenosine, inosine, and hypoxanthine/xanthine measured in tissue and plasma by a luminescence method.

This simple method for sequentially quantifying hypoxanthine (HYP), inosine (INO), and adenosine (ADN) concentrations exploits the H2O2 peroxidase-catalyzed chemiluminescence of luminol. Though applied here only to tissue and plasma, this method can be adapted to analyze other body fluids. HYP in human plasma was stable for 30 min in 10 mmol/L EDTA reagent, whereas ADN was slowly converted to I...
