Muropeptide recycling in Bacillus subtilis:beta-N-acetylglucosaminidase NagZ operates by a unique Asp-His catalytic dyad mechanism
We identified a pathway in the Gram-positive bacterial model organism Bacillus subtilis that is destined for recovery of N-acetylglucosamine-N-acetylmuramic acid-peptides (muroepides) derived from the peptidoglycan of the cell wall. The pathway is encoded by a cluster of six genes of which the first three are orthologs of Escherichia coli genes involved in Nacetylmuramic acid (MurNAc) dissimila...
متن کاملSelective trihydroxyazepane NagZ inhibitors increase sensitivity of Pseudomonas aeruginosa to β-lactams.
AmpC β-lactamase confers resistance to β-lactam antibiotics in many Gram negative bacteria. Inducible expression of AmpC requires an N-acetylglucosaminidase termed NagZ. Here we describe the synthesis and characterization of hydroxyazepane inhibitors of NagZ. We find that these inhibitors enhance the susceptibility of clinically relevant Pseudomonas aeruginosa to β-lactams.
متن کاملThe catalytic mechanism of Pseudomonas aeruginosa cd1 nitrite reductase.
The cd1 NiRs (nitrite reductases) are enzymes catalysing the reduction of nitrite to NO (nitric oxide) in the bacterial energy conversion denitrification process. These enzymes contain two distinct redox centres: one covalently bound c-haem, which is reduced by external electron donors, and another peculiar porphyrin, the d1-haem (3,8-dioxo-17-acrylate-porphyrindione), where nitrite is reduced ...
متن کاملStructure of N-Formimino-l-glutamate Iminohydrolase from Pseudomonas aeruginosa
N-Formimino-l-glutamate iminohydrolase (HutF), from Pseudomonas aeruginosa with a locus tag of Pa5106 ( gi|15600299 ), is a member of the amidohydrolase superfamily. This enzyme catalyzes the deamination of N-formimino-l-glutamate to N-formyl-l-glutamate and ammonia in the histidine degradation pathway. The crystal structure of Pa5106 was determined in the presence of the inhibitors N-formimino...
متن کاملTricarboxylic acid cycle in Pseudomonas aeruginosa.
During the past few years scattered information has appeared which indicates that at least some bacteria probably possess a tricarboxylic acid cycle. Lockwood and Stodola found that Pseudomonas jluorescens formed considerable quantities of a-ketoglutaric acid when growing on a glucose medium under conditions of intense aeration (1). Ajl and Werkman found evidence for at least parts of the conve...
