Canonical Transient Receptor Potential Channel 3 Contributes to Cerebral Blood Flow Changes Associated with Cortical Spreading Depression in Mice

چکیده
Cortical spreading depression is a pathophysiological event shared in migraines, strokes, traumatic brain injuries, and epilepsy. It associated with complex hemodynamic responses, which, turn, contribute to neurological problems. In this study, we investigated the role of canonical transient receptor potential channel 3 (TRPC3) responses elicited by cortical depression. Cerebral blood flow was monitored using laser speckle contrast imaging, triggered three well-established experimental approaches mice. A comparison TRPC3 knockout mice controls revealed that genetic ablation expression significantly altered promoted hyperemia consistently. Our results indicate contributes could be novel therapeutic target for host disorders.

Objective(s): We aimed to investigate the influence of transient receptor potential channel 3 (TRPC3) on lipopolysaccharide-induced (LPS) preterm delivery mice. Materials and Methods: Mice were randomly assigned to the four groups: an unpregnant group, a mid-pregnancy group (E15), a term delivery group, and an LPS-induced preterm delivery group (intraperitoneal injection LPS at 15 days). Uterin...
متن کاملdecreased levels of canonical transient receptor potential channel 3 protein in the rat cerebral cortex after chronic treatment with lithium or valproate
lithium and valproate modulate disturbances in intracellular calcium homeostasis implicated in the pathophysiology of bipolar disorder, but the molecular mechanisms are not fully understood. two subtypes of transient receptor potential (trp) channel family, i.e. trpc3 and trpm2, are potential candidates involved in calcium signaling and implicated in the pathophysiology of bipolar disorder. thi...
متن کاملDiffuse optical correlation tomography of cerebral blood flow during cortical spreading depression in rat brain.
Diffuse optical correlation methods were adapted for three-dimensional (3D) tomography of cerebral blood flow (CBF) in small animal models. The image reconstruction was optimized using a noise model for diffuse correlation tomography which enabled better data selection and regularization. The tomographic approach was demonstrated with simulated data and during in-vivo cortical spreading depress...
متن کاملRelationship between local changes in cortical blood flow and extracellular K+ during spreading depression.
Change of local cerebral blood flow in response to a single cortical spreading depression of Leão was studied in the brain. During a spreading depression, potassium briefly accumulates in the brain extracellular space. The cortical blood flow was normal during the maximal rate of increase of the extracellular potassium concentration. The blood flow doubled during the subsequent period of normal...
