Candidate miRNAs from Oryza sativa for Silencing the Rice Tungro Viruses

Rice tungro disease (RTD), caused by bacilliform virus (RTBV) and spherical (RTSV) is one of the most prominent viral diseases in Asian countries. This problem seems to have been accentuated those countries causing a series outbreaks over years after being first reported International Research Institute (IRRI), Philippines, 1963. One effective ways combat viruses through RNA silencing. microRNA an important player silencing mechanism. Genome sequences analysis shows RTBV-SP isolate (8 Kb) composed four open reading frames (ORF 1, ORF 2, 3, 4), meanwhile, RTSV-SP (12 consists frame encoded seven different polyproteins (P1, CP1, CP2, CP3, NTP, Pro, Rep). Therefore, this study investigated possible rice-encoded miRNAs targeted on RTBV RTSV using silico analysis. Five bioinformatics tools were employed five prediction algorithms: miRanda, RNA22, RNAhybrid, Tapirhybrid, psRNATarget. The results revealed each can be silenced three potentially best candidate miRNA. For RTBV, osa-miR5510 (accession no. MIMAT0022143), osa-miR3980a-3p MIMAT0019676), osa-miR3980b-3p MIMAT0019678) are predicted all algorithms. Meanwhile, for RTSV, osa-miR414 MIMAT0001330), osa-miR5505 MIMAT00221138) osa-miR167a-3p MIMAT0006780). data provide useful material developing RTSV-resistant rice varieties.
