Cancer-associated fibroblasts support bone tropic metastasis by acting as coordinators between the tumor microenvironment and bone matrix in breast cancer

چکیده
Bone is a common site of metastasis for various types cancer cells, including breast cancer, and the consequent skeleton-related events observed in patients are severe often fatal. Currently, it widely accepted that cancer-associated fibroblasts (CAFs) confer metastasis-promoting property to cells. Furthermore, clinical observations suggest CAFs mediate bone tropism metastatic Therefore, deeper understanding mechanism by which involved bone-tropic can facilitate study novel effective therapeutic drugs corresponding targets. In this review, we focused on coordinator role remolding cells remodeling marrow during metastasis. We discussed potential roles CXCL12/CXCR4 axis, CAFs-CSCs reinforcing loop, exosomes malignant process. summary, agreement with Paget's theory, play pivotal colonization providing fertile soil selected seeds influencing tumor-intrinsic characteristics microenvironment (ME).

CCN3 promotes prostate cancer bone metastasis by modulating the tumor-bone microenvironment through RANKL-dependent pathway.
Bone metastasis in patient with advanced-stage prostate cancer, the most commonly diagnosed malignancy in Western countries, increases the risk of intractable bone pain. The nephroblastoma overexpressed (NOV/CCN3) gene, a member of the CCN gene family, is responsible for the secretion of CCN3, a matrix-associated protein involved in many cellular functions. However, the role of CCN3 in prostate...
متن کاملRole of pathologic prognostic factors in breast cancer patients with isolated bone metastasis and relationship between SUVmax and prognostic factors
Introduction: 18F-FDG PET/CT provides very effective results in detecting metastases of breast cancer. In our study, we investigated the relationship between maximum standard uptake value (SUVmax) and prognostic pathologic factors in breast cancer cases with isolated bone metastasis and whether there was any difference in terms of prognostic pathologic factors betwee...
متن کاملInteractions between microenvironment and cancer cells in two animal models of bone metastasis Running title: Bone microenvironment and cancer cells
The preferential proliferation of cancer cells in the bone microenvironment is poorly characterized. Expression pattern of bone marrow and other organs microenvironment in contact with osteolytic (Walker W256) and osteoblastic (MatLyLu MLL) metastases were investigated. Fisher and Copenhagen rats received respectively W256 and MLL cells injection. Bone and soft tissues were analyzed by immunoch...
متن کاملBone Microenvironment and Role of Rank-Rankl-Opg in Breast Cancer Metastasis in Bone
Bone is a frequent site for distant metastasis and more prevalent in breast cancer patients. In advanced breast cancer, more than two thirds of patients will develop high rates of relapse in bone [1,2]. During the bone metastatic phase of breast cancer disease, nearly 50% of patients will be affected by skeletal related events (SRE) including pathologic fracture, bone surgery, spinal cord compr...
متن کاملMatrix metalloproteinase activity, bone matrix turnover, and tumor cell proliferation in prostate cancer bone metastasis.
BACKGROUND The metastasis of prostate cancer to bone is associated with a substantial increase in bone matrix turnover. Matrix metalloproteinases (MMPs) play roles in both normal bone remodeling and invasion and metastasis of prostate cancer. This study was designed to determine the role of MMP activity in prostate cancer that has metastasized to bone. METHODS Single human fetal bone fragment...
