BLOC1S1: An Unexpected Regulator of Th2 Cell-driven Inflammatory Responses

چکیده

Abstract BLOC1S1 is an adaptor protein that modulates endosome-lysosome function as a component of numerous multiprotein complexes. We postulated its modulation in CD4 +T cells may help us uncover the role biology adaptive immunity. In primary cre-recombinase knockout resulted preferential induction Th2 compared to Th1 and Th17 cytokine production following T cell receptor activation. As atopic dermatitis allergic inflammatory skin disease characterized by type 2 cytokines we evaluated effect KO vs controls calcipotriol-induced mice. The absence exhibited exacerbated accompanied with significantly elevated plasma IgE, eosinophilia, responses, pathology relative wild-type transcription factor Gata-3 expressed but not plays critical differentiation dermatitis. deficient showed enhanced nuclear kB (NF-kB), Gata3 LC3 positive autophagosomes. Inhibition NF-kB activity prevented expression, autophagosomes −/− cells. This mouse model will now give platform explore endo-lysosomal polarization NHLBI Division Intramural Research (MNS-ZIA-HL005102)

Cd8+ T-Cell Responses in a Th2-Polarized Inflammatory Setting

A variety of external factors can affect the activation status, effector function, and memory differentiation of T cells. Such factors include resident microbiota and the cytokine milieu produced in a host. Loss of tolerance to commensal bacteria and excess inflammatory cytokines such as IL-4 have been implicated in the pathogenesis of many diseases, including inflammatory bowel disease (IBD) a...

Th2 and eosinophil responses suppress inflammatory arthritis

Th2-eosinophil immune responses are well known for mediating host defence against helminths. Herein we describe a function of Th2-eosinophil responses in counteracting the development of arthritis. In two independent models of arthritis, Nippostrongylus brasiliensis infection leads to Th2 and eosinophil accumulation in the joints associated with robust inhibition of arthritis and protection fro...

RGS16 attenuates pulmonary Th2/Th17 inflammatory responses.

The regulators of G protein signaling (RGS) protein superfamily negatively controls G protein-coupled receptor signal transduction pathways. RGS16 is enriched in activated/effector T lymphocytes. In this paper, we show that RGS16 constrains pulmonary inflammation by regulating chemokine-induced T cell trafficking in response to challenge with Schistosoma mansoni. Naive Rgs16(-/-) mice were "pri...

Epithelial reticulon 4B (Nogo-B) is an endogenous regulator of Th2-driven lung inflammation

Nogo-B is a member of the reticulon family of proteins (RTN-4B) that is highly expressed in lung tissue; however, its function remains unknown. We show that mice with Th2-driven lung inflammation results in a loss of Nogo expression in airway epithelium and smooth muscle compared with nonallergic mice, a finding which is replicated in severe human asthma. Mice lacking Nogo-A/B (Nogo-KO) display...

WTX: an unexpected regulator for p53.

The WTX gene is frequently lost or mutated in Wilms tumor. In this issue of Molecular Cell, Kim et al. (2012) identify WTX modulation of p53 tumor-suppressor activity through regulation of p53 acetylation. Therefore, WTX differentially regulates the oncogenic β-catenin pathway and the tumor-suppressing p53 pathway.
